With the development of endodontology and material science, in recent years, appeared many new product root canal sealer, its good liquidity is one of the factors to ensure that root canal filling closely. Clinical often see some accepted norms of root canal treatment failure cases, this is in addition to related to the complexity of the root canal system, and root canal sealer lysate may also become a hapten impact on periapical tissue. In clinical application there are many kinds of root canal sealer be used by dentist without knowing its biological safety well. Therefore, we selected the6kinds of new type of root canal sealer in vitro cytotoxicity experiments, hope it played a guiding role for clinical applications.Objective: To detect the cytotoxic effects of AH plus, Adseal,2Seal, Reoko seal, EZ-fill and iRoot SP on L-929cell line.Methods:In this in vitro experimental study, the materials were mixed according to the manufacturers’instructions. The elutes of materials were prepared separately and were incubated with L-929cell line for24hour,48hours,and72hours. Cell viability was evaluated by MTT assay.Results:Within the same group of incubation time, all the OD value of the leaching solution with less increase of leaching time (P<0.05).Within the same set of leaching time, the OD value of the maximum value appeared in co-culture48h;24h and48h OD value of the significant difference (P<0.05); the OD value of the leaching solution is slightly lower in co-culture72h, there is no significant difference between48h OD value and72h OD value (P>0.05).Conclusions:The cytotoxic of root canal sealer increases with leaching time. Cell toxicity comparison of6kinds of root canal sealer: RoekoSeal no cytotoxicity, EZ fill and2seal are better than AH plus, Adseal and iRoot SP. Under the influence of experimental conditions, the cytotoxicity evaluation of iRoot SP with less sense.
