Group A was made byusing ligating4cm-long ileum for1h and group B was for3h. Whenintestinal obstruction models were built, the changes of pathology in liverand intestine tissue were observed by light microscope. The Park/Chiuscale was used to evaluate the severity of intestinal lesion. Serumconcentration of HDC and D-lactate were determined by ELISA. RT-PCRand real time quantitative PCR were used to detect the expression ofHDC in liver and intestinal tissue. Results: Under light microscopic level, the injury score of intestinein test group was increased in comparison with that in control group. At1h and3h after strangulated intestinal obstruction, concentration of serumHDC were83.9727.69pg/ml and143.2147.15pg/ml respectively, which were significantly higher than those in controlgroup(9.973.64pg/ml, P<0.01). At3h after strangulated intestinalobstruction, concentration of serum D-lactate was increased to10.3614.91ng/ml, but this change had no statistical significancecompared with the control(1.111.79ng/ml, P>0.05). At3h after strangulated intestinal obstruction, RT-PCR showed that brightness ofHDC band in intestine tissue was significantly higher than the control.Real time quantitative PCR also showed that the expression of HDCmRNA in liver tissue in group A and group B increased1.34fold(P>0.05)and2.21fold(P<0.05) respectively, when compared with the controlgroup. The expression of HDC mRNA in intestine tissue in group A andgroup B increased1.81fold(P>0.05) and8.02fold(P<0.01) respectively, when compared with the control group.Conclusion:1.HDC mRNA expressed more in liver and intestinetissue in strangulated intestinal obstruction.2.Concentration of serumHDC level was significantly increased in early strangulated intestinalobstruction.3.Concentration of serum D-lactate had no significant changein early stage of small scale intestinal ischemia.4.Expression of HDCcan be used for early diagnosis of strangulated intestinal obstruction.5.High expression of HDC may indicate the severity of intestinalobstruction. | | Keywords/Search Tags: | Histidine decarboxylase, D-lactate, Strangulated intestinalobstruction, Early diagnosis | PDF Full Text Request | Related items |
