[Objective]To detective the feasibility of simulating spleen deficiency syndrome of STC by the way of sennae water restriction and diet control, and detective the theory of zhizhuwan regulate the intestinal movement by observing the in fluence of zhizhuwan on expression of AchE、5-HT7R and AQPs in colon tissue of mice with spleen deficiency syndrome of STC.[Methods]Fist, simulating the model of spleen deficiency syndrome of STC:Using80SPF Kunming mice to divided into the contron group (20) and the model group (60), the first week of the model group lavaging the sennae20g/(kg.d) and1kg/ml, Then stop lavaging sennae at the eighth day and only feeding rice4-8g and drinking water once for0.5h in every two days for8days, After the success of modeling, divided the model group into zhizhuwan high dose group mideal dose group marenwan group and mosapride group, and lavaging at the sixteenth day for7days, then testing the general behavior characteristics, first black defecating time, the defecation quantity and quality in6h, the serum xylose concentration and the characteristics of the colonic mucosa tissue pathology; Testing the expression of AchE、5-HT7R and AQPs by Immunohistochemistry and the expression of5-HT7R and AQPs mRNA by Real-time PCR.[Results]1.Compared with the model group, the weight of the control group and each treating group were significantly increased(P<0.01).2.the Observation of defecation:compared with the control group, the first black defecating time was significantly increased and the other treating group were more significantly decreased than the model group, and the zhizhuwan middle dose group was most significant(P<0.01), and the competing between zhizhuwan high dose group and marenwan group were nonsignificant(P>0.05); compared with the mosapride group, other treating group were significantly decreased(P<0.01).3. The serum xylose-D concentration:compared with the control group, the serum xylose-D concentration of model group was significantly decreased, and compared with the model group, other treating group but mosapride group were significantly increased(P<0.01)., and the mosapride group was nonsignificantly increased(P>0.05).4. Immunohistochemistry and Real-time PCR testing:compared with the control group, the concentration of AchE of the model group was significantly decreased and5-HT7R、AQP4、AQP8were significantly increased, and the zhizhuwan middle dose group was more significant increased and5-HT7R、AQP4、AQP8were more significantly increased than other treating group(P<0.01).[Conclusions]1. Spleen deficiency syndrome of STC exsists the abnormal expression of Enteric neurotransmitters and AQPs, as decreasing the expression of AchE and increasing the expression of5-HT7R in colon tissue.2. zhizhuwan can significantly improve the function of spleen catharsis to spleen deficiency syndrome of STC3.zhizhuwan could improve the expression of of AchE in colon tissue of spleen deficiency syndrome of STC mice, and improve the intestine movement by decreasing the expression of5-HT7R, and decrease the colon on water absorption by deceasing the expression of AQP8、AQP4to improve the fecal water content to treat constipation.4.zhizhuwan maybe treat spleen deficiency syndrome of STC by regulating the Enteric neurotransmitters (AchE、5-HT7R) and the expression of water channel protein (AQP8、AQP4).
