Objective:To investigate the effect of the piper nigrum’s active constituent5,7-dimethoxyflavone(DMF) on invasion^metastasis and expression of Snail protein in ovarian cancer sphere forming cells (OCSLCs) derived from human ovarian cancer SKOV3cell line, and preliminary discussing on whether the effect of piper nigrum’s active constituent (DMF) can inhibit OCSLCs invasion,metastasis and the protein expression of Snail,and to observe the OCSLCs invasion,metastasis whether associated with the regulation of q proteinwas. so as to provide a new approach for the treatment of ovarian cancer.Methods:Human ovarian cancer was cultured in vitro. Used the suspension culture with stem cell-conditioned medium to enrich and gather the Ovarian cancer sphere-forming cells and identified as OCSLCs. the ability of cell invasion and metastasis were investigated in human ovarian cancer SKOV3cell line and OCSLCs using Transwell assay.And the Effect of Piper nigrum’s active constituent (DMF) on its invasion and metastasis.We used the Western blot to compare the expression of Twist protein in SKOV3sphere-forming cells and parental SKOV3cells, and the effect of piper nigrum’s active constituent (DMF) on expression of Snail protein in OCSLCs.Results:1. The ovarian cancer sphere cells derived from human ovarian cancer SKOV3cell line which was cultured in stem cell-conditioned medium condition growing into a sphere.2.The Transwell assay showed that OCSLCs with high invasion, high metastasis.3. Piper nigrum’s active constituent (DMF) could significantly inhibit OCSLCs cell invasion and migration rate.4.Piper nigrum’s active constituent (DMF) could inhibit expression of Snail protein with the dose-dependent manner.Conclusion:1. Ovarian cancer sphere-forming cells derived from ovarian cancer SKOV3cells which was cultured in stem cell-conditioned medium have high invasion、high metastasis characteristics.2. Piper nigrum’s active constituent (DMF) can inhibit high invasion、high metastasis ability of OCSLCs.3. The invasion and metastasis inhibited role of piper nigrum’s active constituent (DMF) of OCSLCs were associated with down-regulating expression cancer signaling molecule Snail.4. Piper nigrum’s active constituent (DMF) inhibited the invasion and metastasis abilities of OCSLCs.and regulated the expression level of Snail protein; it maybe a potential approach for curing the ovarian cancer focusing on the key pathological factors "virtual"," cold","phlegm"," blood stasis" and "toxin"...
