Objective:to study the influence of Ganlu Xiaodu Decoction over the enzymatic activity of neonatal mice infected with EV71virus with the purpose of providing experimental basis for the explanation on the efficacy mechanism of Ganlu Xiaodu Decoction.Method:The7-day-old mice was subcutaneously vaccinated with20μl100LD50of EV71virus to build a mice’s EV71infection model. After the modeling Ganlu Xiaodu Decoction fed to the wholeparty and demolished party and Ribavirin treatment。The normal group and model group were given sterile water of the same amount by way of gavage, and after10d treatment, the neonatal mice was killed for the purpose of testing the enzymatic changes.Results:After observation for14days, Changes in serum enzyme Ganlu Xiaodu Decoction and the other six serum enzymes exist significant differences (P<0.05).After treatment with Ganlu Xiaodu Dan, serum enzyme levels in mice decreased gradually. Treatment of pediatric hand-foot-mouth disease (HFMD) with combined Ganlu Xiaodu Dan and ribavirin showed marked effects on myelin basic protease (MBP), lactate dehydrogenase (LDH), creatine kinase (CK) and neurone specific enolase (NSE)(P<0.05). Analysis results suggest that Ganlu Xiaodu Dan has an evident anti-EV71activity, plays an important role in the efficacy of pediatric HFMD, and has remarkable effects in treatment of myocardium, brain and central nervous system in severe pediatric HFMD. Ganlu Xiaodu Decoction decomposed recipe on the treatment of children with hand, foot and mouth disease also have a therapeutic effect, but the treatment is less than willing to all parties. Ribavirin, on the other hand, had an obvious effect on NSE; whose level decreased gradually on7th,9th and12th days, and exhibited a gradual upward trend on the14th day. Other pairwise enzymatic comparisons showed statistically significant differences (P<0.05). Chinese and western medicines both had a certain efficacy for pediatric HFMD, but they had both similarities and differences in terms of regulatory effect. Ganlu Xiaodu Dan decoction had a more pronounced regulatory effect on serum enzymes than ribavirin.Conclusion:1.Ganlu Xiaodu Dan can reduce the MBP, LDH, CK and NSE levels in sera of EV71infected-neonatal mice.2.Effects of Ganlu Xiaodu Dan on serum LDH, NSE, MBP and CK of EV71infected-neonatal mice on8th and10th days after treatment change markedly.3.Combined Ganlu Xiaodu Dan and ribavirin may have a protective effect on tissue cells of EV71infected-neonatal mice by reducing MBP, LDH, CK and NSE levels.
