We not only studied the pharmacodynamics of Periplaneta americana against hepatic injury caused by CCl4, but also the preparation and quality evaluation of its extract with water (SEW).First, the drug effect of its powder and extracts was investigated by in vivo experiment respectively; then the extracts were compared from the molecular weight and content of protein; after that the products of EW separated by gradient ammonium sulfate fractionation method was tested in order to screen the active fraction; The results showed that its powder had poor efficacy, as well as the extracts with alcohol (EA) and the refluxing extract with water (REW), while the efficacy of SEW was satisfactory, and the electrophoresis map of SEW was quite different from the one of EA and REW, and the former had the highest protein content.Secondly, the preparation of its extract with water (EW) was optimized:the optimizing extraction parameters that were obtained by single factor test and orthogonal design were as follow:P.americana coarse powder was extracted with25times the amount of water at70V and55℃for i time,3.5minutes every time; the study on the influence of crude drug content on purification effect showed that the optimal crude drug content was0.5g·mL-1; the EW was dried by vacuum drying at60℃. Finally, the quality study of EW included character identification, physical and chemical identification, the percentage of containing water and content determination. With hygroscopicity, bulk density and angle of repose as indexes, several factors such as the kind, ratio and dosage of fillers were investigated with single factor test, and optimum concentration of wetting agent was screening with difficulty degree of granulation and granule character as indicators. The optimum formulation process was:with90%alcohol as wetting agent, EW, soluble starch and microcrystalline cellulose mixed by50:25:25was granulated by extruding granulation, after being dried the granules were filled into0#empty capsules (each one weighed0.29g). The quality study of P. americana capsule was base on the one of EW combining with capsule quality control.According to this research, the product of P. americana EW salted out at25-50%ammonium sulfate saturation is effective; and the preparation process above is reasonable, the quality of this preparation is stable and controllable.
