| Purpose:The concept "the liver open into the eyes" is a component of the"zang-organs and orifices theory" in TCM.But now,the biological mechanism for the"the liver open into the eyes" concept is not clear enough.In this study, we searchedmainly by animal experiments,cut into the point of biological mechanisms,andexpression mechanism for research liver depression at road N-methy-D-aspartatereceptor one(Referred to as NMDAR1or NR1) expression and the effect of explorethe biological mechanism of eye and liver correlation,interpretation theory of "liverbegin to understand the eye",the theory of "the liver open into the eyes" to guide theclinical practice.Method:(1)Theoretical Study:We consulted the ancient literature and the modernliterature to tease out the origin about the theory of "the liver and the eyes",searchedin the relevant literature on the international journals about "the liver and the eyes", tointerpretation the relationship between the eye and liver progress andtrends.(2)Experimental Study: we choosed to change for the interference factors inthe relationship between the eye and liver, chronic stress method is used to copy theliver depression syndrome rats models, observed the liver pathogenesis change, theimpact on purpose.Select the liver on behalf of the formula at Xiaoyaosanintervention, to observe treated from liver,impact on purpose.From the level of organ observation of the retina and visual cortex,lateral geniculate body nerve cellmorphological changes, from the molecular level observation retinal nitricoxide(NO)〠malondialdehyde(MDA),observed from protein and gene levelsdepending on the road covering cortex,lateral geniculate body spatial and temporalvariations of NR1,in order to explore the relationship between eye and livermechanism in the middle,to investigate the intermediate mechanism of therelationship between the liver and the eyes.Results: After the stimulation in rats by chronic stress modeling threeweeks,compared with the normal group, the model group decreased appetite and lostweight, depressed, listlessness;index of sucrose preference was significantly lower;inOpen-field test,the residence time of the central grid increased, level crossing and thenumber of cells,the number of standing,modification times significantly reduced;thelevels of plasma ACTH elevated;The different degrees of organic disease occurred inliver;In immunohistochemical staining,the protein expression of NMDAR1in thevisual cortex and the lateral geniculate body increased;In situ hybridizationstaining,the gene expression of NR1mRNA in the visual cortex and the lateralgeniculate body significantly increased.But compared with the model group,the rat ofmedium-dose and high-dose Xiaoyaosan group in immunohistochemical staining,theprotein expression of NMDAR1in the visual cortex and the lateral geniculate bodydecreased;In situ hybridization staining,the gene expression of NR1mRNA in thevisual cortex and the lateral geniculate body significantly decreased.Conclusion:This experiment used by chronic stress by liver depression syndromerats model was a success.Liver depression syndrome rats NR1higher protein andgene expression, internal factors may be liver depression syndrome affect purpose,and depending on the road damage mechanism is discussed in this paper related toglutamate receptor excitability toxicity;At the same time, the liver xiaoyao powder the formulas for eye function to improve the effect of liver depression syndrome rats,counterevidence liver depression effect on purpose.Further illustrate NR1can be usedas to explore the theory of "liver begin to understand the item" biologicalmechanisms. |
