Objective:Through experiment to confirm the correlation between fever-related factors with "nian" nature brain hurt,"nian" nature heat syndrome during the transformation from immature to mature period in rabbit models. Through applying modern science to elaborate the pathogenesis change rule of Mongolian medicine warm disease, which provides an experimental basis for dialectical diagnosis and treatment of Mongolian medicine heat syndrome (infectious autoimmune diseases).Methods:Two kinds of pathogens were injected into rabbits by different ways to establish the animal models of "nian" nature brain hurt and "nian" nature heat syndrome of immature and mature period. The cerebrospinal fluid and peripheral blood were collected at null point (before injection), immature and mature heat period, and the IL-1, IL-6and TNF-α levels in the samples collected three times were determined by ELISA.Modeling methods:(1)"Nian" nature rabbit brain hurt model was established by cisterna magna injection of Neisseria meningitidis. Each rabbit in the model group was injected3×108cfu/ml Neisseria meningitidis solution into cisterna magna; each rabbit in the control group was injected the same amount of normal saline. Every20minutes after injection, the body temperature was measured and the syndromes manifestation was observed, and during this period the rabbits diet and activities were normal. The same amount of peripheral blood and cerebrospinal fluid at null point were extracted. The peripheral blood and cerebrospinal fluid at immature and mature heat stage were collected after injection1.5to2.5hours and3.5to4.5hours respectively.(2)"Nian" nature rabbit fever model was established by ear vein injection of Escherichia coli. Each rabbit in the model group was injected8×107cells/ml of E.coli dilution into ear vein, and each rabbit in the control group was injected the same amount of normal saline. The same amount of peripheral blood at null point was extracted, the body temperature was measured and the syndromes performance was observed. The peripheral blood at immature and mature heat stage was collected after injection2.5to3.5hours and5to6hours respectively.Results:1.Brain hurt group:from the view of heating process, it possesses the fervescence, hyperpyrexia and defervescence stage, which not only conforms to the fever process of human diseases, but also conforms to the transformation process from immature to mature heat period of Mongolian medicine warm disease. From the view of fever degree, it was consistent with the clinical manifestations and diagnostic criteria of Mongolian medicine warm disease, so the brain hurt model was successfully established. The IL-1and IL-6content in cerebrospinal fluid:compared with the control group, the immature and mature heat period in the model group both were significantly different (P<0.05); in the model group, compared with null point, the immature and mature heat period both were significantly different (P<0.05). TNF-a content:compared with the control group, the immature and mature heat period in the model group both were significantly different (P<0.05); in the model group, compared with null point, the immature and mature heat period both were significantly different (P<0.05). and there was a significant difference between immature and mature heat period (P<0.05). WBC in cerebrospinal fluid:Compared with the control group, the mature heat period in the model group was significantly different (P<0.05):in the model group, compared with null point, the immature and mature heat period both were significantly differene (P<0.01):but there was no significant difference (P>0.05) between immature and mature heat period. CRP content in cerebrospinal fluid:the immature and mature heat period between control and model group, and within model group were not significantly different (P>0.05).2."Nian" nature heat group:the heat process and heat syndromes of model group meet the Mongolian medicine transformation process from immature to mature period, indicating the "nian" nature fever model was successfully established. The IL-1. IL-6levels in peripheral blood:compared with the control group, the immature and mature heat stage in the model group both were significantly different (P<0.05):in the model group, compared with null point, the immature and mature heat period both were significantly different (P<0.05). TNF-α content:compared with the control group, the immature and mature heat period in the model group both were significantly different (P<0.05); in the model group, compared with null point, the immature and mature heat period both were significantly different (P<0.05). and there was a significant difference between immature and mature heat period (P<0.05). Peripheral WBC: compared with the control group, the mature heat period in the model group was significantly different (P<0.05); in the model group, compared with null point, the immature and mature heat period both were significantly different (P<0.01), and there was a significant difference between immature and mature heat period (P<0.05). CRP levels:the immature and mature heat period between control and model group, and within model group were no significant difference (P>0.05).Conelusion:1. Application of cisterna magna injection of Neisseria meningitidis and ear vein injection of Escherichia coli successfully establish "nian" nature brain hurt syndrome model and "nian" nature heat syndrome model of transformation from immature to mature heat syndrome respectively,that accord with the clinical syndromes manifestation of human being, and confirm the diagnostic criteria, which is advanced by prophase research of the transformation from immature to mature period in rabbit models.2. The two model groups both conform to the theory of Mongolian medicine, and can be used to study the pathogenesis of Mongolian medicine febrile disease and relevance with it.3. The two model groups both are confirm that the change of the transformation from immature to mature has certain relevance with WBC number and IL-1, IL-6and TNF-α level.4. The vary temperature curve, fever time, symptom and symptom persistence time of heat syndrome were induce by different germ and ways of administer dosage.
