Objective: To analysis the frequencies distribution of SNPs in ABCB1C1236T andABCA1R1587K in Anhui Han dyslipidemia and ortholiposis crowd and the correlationbetween these SNPs and lipid levels.To investigate the correlation between SNPs ofABCB1C1236T and ABCA1R1587K and lipid-lowering efficacy of atorvastatin.Methods: The polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) method was used to determine the SNPs of ABCB1C1236T and ABCA1R1587K from197dyslipidemia patients and26ortholiposis subjects.114cases withdyslipidemia were treated with20mg atorvastatin every night and fasting lipid levelswere measured before and after one month of treatment.Results: The mutation frequencies of ABCB1C1236T and ABCA1R1587K allele inAnhui Han dyslipidemia crowd were69.54%and60.41%and in ortholiposis crowdwere53.85%'69.23%.It was different between dyslipidemia and ortholiposis crowd inABCB1C1236T genetype distribution(P<0.05)and it was not different betweendyslipidemia and ortholiposis crowd in ABCA1R1587K genotype distribution(P>0.05).The distribution of ABCB1C1236T and ABCA1R1587K genotype and allelewere consistent with Hardy-Weinberg equilibrium.In197dyslipidemia patients,ABCB1C1236T CC genotype had higher plasma LP(α)levels than CT genotype(P<0.05)andTT genotype(P<0.05)and ABCA1R1587K AA genotype had lower plasma APOAIlevels than AG genotype(P<0.01)and GG genotype(P<0.05).It could decrease plasma TC、TG、LDL-C、APOB levels and increase plasma HDL-C、APOAI、LP(α)levels after treated with20mg atorvastatin one month.There were98cases had completechinical data in114dyslipidemia patients treated with atorvastatin,ABCA1R1587K AGgenotype had higher plasma HDL-C levels than GG genotype(P<0.05)and plasmaAPOAI levels in AA genotype、AG genotype and GG genotype showed significantdifferences(p>0.05) before treated with atorvastatin.ABCB1C1236T CC genotype hadhigher plasma LP(α) levels than TT genotype(P<0.01)and ABCA1R1587K GGgenotype had higher plasma LP(α) levels than AA genotype(P<0.05)and AG genotype(P<0.05)after treated with atorvastatin. The change rate of plasmaAPOAI levels inABCA1R1587K AA genotype was higher than GG genotype(P=0.01).Conclusion:1.ABCB1C1236T genotype distribution in dyslipidemia crowd is differentfrom ortholiposis crowd.2.There is a correlation between ABCB1C1236T and ABCA1R1587K SNPs and plasma lipid levels.3.It can obtain satisfactory lipid-loweringefficacy taking20mg atorvastatin.4.ABCB1C1236T SNPs have no relationship withlipid-lowering efficacy of atorvastatin.5.ABCA1R1587K SNPs have a relationship withlipid-lowering efficacy of atorvastatin.
