BackgroundOvarian cancer is one of the most popular gynaecologic oncology. As its strong malignant behavior and tend to planting in Abdomen, Ovarian cancer has the highest recurrence rate and mortality among gynaecologic malignancy oncology. LIMK-1, one of the important molecules that regulate structure of cytoskeletal, plays a vital role in tumor cell proliferation and invasion. MicroRNAs are non-coding RNAs, which have been shown to play crucial roles in diverse biological processes, such as cell development, signal transduction, tissue differentiation and maintenance, disease, and carcinogenesis. Although the study we have done previously shows that miR-126has the ability to modulate tumor cell proliferation and invasion by regulating the expression of VEGF, the mechanism is still unclear. In this study, by means of culturing SKOV3ovarian cancer cells in vitro, and transfected with miR-126mimics, we attempt to indicate the signaling pathway of VEGF/LIMK-1in ovarian cancer invasion and metastasis.Materials and MethodsThe ovarian cancer cell line used in this study was SKOV3, transfected with miR-126mimics. The control team included LV3NC and LV3-has-miR-126transfected cells. By immunofluorescence, we compared the difference of LIMK-1expression in each group and analysed the relationship of miR-126and LIMK-1. AndTranswell invasion assay was carried out to observe the invasion ability of each group, then to analyze the relationship of expression of miR-126and LIMK-1in ovarian cancer cell invasion ability..Results1.261±8cells invaded the matrigel-coated membrane in the SKOV3group,276±20(P>0.05) in the controlling team, and232±17(P<0.05) in the LV3-has-miR-126group amid the cell invasion teams.2. The proportion of LIMK-1protein expression was almost the same comparing the SKOV3group to the SKOV3/LV3NC group. However, LV3-has-miR-126group was significantly lower than the other groups.Conclusion1. The result of Transwell assay gave a hint that expression of miR-126played a role in invasion of ovarian cancer.2. Expression of LIMK-1was decreased in cells transfected with miR-126,mimics which revealed expression of LIMK-1may related to the effect.
