Objective:In the recent years, the incidence of salpingitis is more and more prevalent, which severely affects human reproduction health. How to effectively treat and prevent the sequela of salpingitis is the problem need to be solved. As the first line of defense against pathogens infection, Toll-like receptors is one of the most important natural immunity receptor family. The appropriate expression of Toll-like receptors was beneficial to eliminating pathogens, however, the excessive or insufficient express of Toll-like receptors may lead to the occurrence of the oviduct inflammatory. TLR2and TLR4expression in the oviduct tissue, and can be combined with pathogen-associated molecular patterns of staphylococcus aureus involved in the formation of salpingitis. It is traditionally considered that glucocorticoids (GCs) can suppress immune response, weaken the body’s resistance to infection, but GCs used in the clinical treatment has not been universally accepted. Even though the use of antibiotics could solve the problem of tubal infection, however, antibiotics can not solve the problem of tissue damage to the fallopian tubes because of inflammatory. In the paper, we studied the effect of the low, medium, and high-dose doses of dexamethasone (DXM) intervention on the expression of TLR2mRNA and TLR4mRNA in the tissues of the mice oviduct with staphylococcus aureus inflammation. Furthermore, we studied the regulation of inflammatory responses to different concentrations of DXM in order to understand its impact on healing after tubal inflammation, thus improving the prognosis of tubal inflammation and fertility outcome.Methods:35mice were randomly divided into normal blank group, staphylococcus aureus group,1mg/kg DXM group,5mg/kg DXM group,10mg/kg DXM groups, Penicillin group,1mg/kg DXM+Penicillin group.5mice in each group. Thus, Staphylococcus aureus-induced mice salpingitis model was established, blank group: no treatment; staphylococcus aureus group:vaginally injection of staphylococcus aureus(1×109/mL,50ul).1mg/kg DXM group:vaginally injection methond same to staphylococcus aureus group, intraperitoneally injection of1mg/kgDXM after3days, daily once for four consecutive days.5mg/kg DXM group:vaginally injection methond same to staphylococcus aureus group, intraperitoneally injection of5mg/kgDXM after3days, daily once for four consecutive days. lOmg/kg DXM groups:vaginally injection methond same to staphylococcus aureus group, intraperitoneally injection of10mg/kgDXM after3days, daily once for four consecutive days. Penicillin group:vaginally injection methond same to staphylococcus aureus group, intraperitoneally injection of50000units Penicillin after3days, daily once for four consecutive days.1mg/kg DXM+Penicillin group: vaginally injection methond same to staphylococcus aureus group, intraperitoneally injection of1mg/kg DXM and50000units Penicillin after3days, daily once for four consecutive days. The expression of TLR2mRNA and TLR4mRNA in mouse oviduct tissue were measured by Real-time PCR. Criteria to determine the degree of inflammation tubal pathology:"-" Indicates that the epithelial cells arranged in neat rows, no substantial degeneration, necrosis, cilia-rich, luminal stenosis, no connective tissue hyperplasia. inflammatory cell infiltration;"+" Indicates that the epithelial cells were cuboidal or low columnar, ciliated thickening、some disappeared, Lumen of a small amount of adhesions, a small amount of connective tissue hyperplasia, a small amount of inflammatory cell infiltration;"++" indicates that the epithelial cells were flat, further loss of cilia, Luminal portion of adhesions, proliferation of the connective tissue part, some infiltration of inflammatory cells;"+++" indicates that a large number of epithelial cell proliferation, lumen severe adhesions, cilia completely disappeared, a significant proliferation of connective tissue, A large number of inflammatory cells infiltration.Results:1. Pathological observation blank group:tubal wall organizational structure clear, luminal patency, mucosal layer of the visible rows of epithelial cells, rich in cilia. Except DXM1+penicillin group, the other experimental groups lumen varying degrees of thickening, severe adhesions, cilia varying degrees of loss or even disappear, connective tissue hyperplasia, inflammatory cell infiltration.. This experiment tubal lesion degree of groups of mice shows that:10mg/kgDXM group>5mg/kgDXM group>1mg/kgDXM group> staphylococcus aureus group>Penicillin group>1mg/kgDXM+Penicillin group.2. Rt-pcr to detect TLR2mRNA and TLR4mRNA expression in salpingitis organizations have varying degrees of increase.①TLR2mRNA expression: staphylococcus aureus group was significantly higher than the blank group (p<0.05),1,5,10mg/kgDXM group and Staphylococcus aureus group reached statistical significance (p<0.05), With the reduction in the dose of DXM, increased expression,1mg/kgDXM set the highest expression. TLR2mRNA expression is:1mg/kgDXM> penicillin group>5mg/kgDXM group> staphylococcus group>10mg/kgDXM group>1mg/kgDXM+penicillin group> blank group.②the expression of TLR4mRNA:staphylococcus group group expression slightly than blank group, no statistical significance (p>0.05). Highest expression in1mg/kgDXM group, penicillin group compared with Staphylococcus aureus group reached statistical significance (p <0.05)1mg/kgDXM group、1mg/kgDXM+penicillin group and Staphylococcus aureus group showed no significant difference (p>0.05). TLR4mRNA expression quantity characterized by:1mg/kgDXM group>5mg/kgDXM group>1mg/kgDXM+penicillin group> staphylococcus group> blank group>10mg/kgDXM group> penicillin group.Conclusion:1. Regulation the expression of TLR4mRNA TLR2mRNA may be one of the factors dexamethasone treatment of salpingitis.2. Penicillin combined with low dose DXM can improve the prognosis of tubal inflammation.
