Purpose:To evaluate the killing effect of neutrophil extracellular traps (NETs) on Staphylococcus aureus and explore the influence of different concentrations of Glucocorticoids on the formation of NETs and the killing efficiency.Methods:First, isolated neutrophils from the whole blood of healthy volunteers; Second, adding a certains concentration of Staphylococcus aureus to neutrophils,observe the formation of NETs as well as the characteristics via immunofluorescence staining,measure efficiency of killing; Then, three different concentrations of dexamethasone(1,10,100μg/ml) were applied to study the influence of dexamethasone on the formation of NETs and its potential effect on neutrophils’efficiency of killing bacteria. Results:Neutrophils in vitro could not only show the activity of phagocytosis, but also display the typical characteristics of NETs.NETs formation rate have a positive correlation with the duration of action, the killing efficiency was positively correlated within two hours;the killing efficiency decreased significantly after DNase hydrolysis NETs。NETs formation and the killing efficiency is affected by dexamethasone, high concentrations of dexamethasone suppression of NETs formation and the killing efficiency.Conclusions:Staphylococcus aureus induces neutrophil formation NETs, The phenomena of phagocytosis and NETs coexist in the anti-infective process of neutrophils;NETs have superior antibacterial function.NETs formation and the killing efficiency was positively correlated within a certain time; glucocorticoids may inhibit the formation of NETs thereby inhibiting neutrophil anti-inflammatory effects. |
