Objective:To investigate the activation status of X chromosomes in complete hydatidiform mole with46,XX karyotype.Method:qRT-PCR was performed to determine the RNA expression of XIST and TSIX that are related to X chromosome inactivation in complete hydatidiform mole and normal villi with46, XX karyotype. Additionally, qRT-PCR and immunohistochemical staining were performed to assay the expression of X-linked gene CDX4in complete hydatidiform mole and normal villi with46, XX karyotype.Results:1. The XIST RNA expression was lower and the TSIX RNA expression was higher in complete hydatidiform mole compared with normal villi with46, XX karyotype.2. The expressions of CDX4mRNA and protein were elevated in complete hydatidiform mole compared with normal villi with46, XX karyotype.Conclusion:Both X chromosomes are activated in complete hydatidiform mole with46, XX karyotype. |
