ObjectivePsoriasis is a common and chronic skin disease which is recurrent, The etiology and pathogenesis have not been yet clear,but it is clear that the interaction of gene and environment is the reason of it. With the rapid development of human gene group plans and the emerging of genetic technologies, psoriasis-related gene location and identification obtain the support of technology and methodology.This study extracted the mRNA from skin tissue of psoriasis patients and normal people then did expression profiling analysis in order to find out the different genes,through analying the characters and fuctions of the different genes,we can make futher discussion on the cause of psoriasis.MethodsIn the study,we chose3patients with psoriasis vulgaris and3normal people, then removed a little tissue from each person,(we removed the lesions of patients),then extracted and evaluated the qulity of total RNA.Purifying the total mRNA and reversely transcripte to single-stranded cDNA,then double-stranded cDNA,purified the double-stranded cDNA and biotin-labled the DNA and transcripted to cRNA.The obtained target gene were hybridized to6Affemetrix gene chip.After being stained and washed,the hybridized chips were scanned by Affymetrix GeneChip Scanner3000,then use Affymetrix GeneChip Operating Software Versionl.4to reading and making data processing.Choosing the different genes according to q-value(%)=5and fold chang=2.Resultsâ‘ There were1592common different genes in skin tissue between psoriasis patients and normal people.774genes were upragulated (account for48.62%),818genes were downregulated (account for51.38%),these differently expressed genes were involved in the biological processes such as inflammation, epidermis development, ketatinocy differentiation, anti-apoptosis, cell adhesion, chemotaxis, immune response,positive regulation of cell proliferation.â‘¡we put genes which fold change>2.5into MAS system and analied deta with Pathway and GO,the results showed that:these different genes were involved in some signalling pathway such as p53,JAK-STAT,Toll-liked receptor,apotosis, cyclins and cell cycle regulation and some signalling pathway through cytokins.Some of them were consistent with the previous studies.ConclusionIn summary, Psoriasis is a genetic disease which is related with multiple genes,the psoriasis is not led by the mutation of single gene but by many gene groups which were interacted by each other and form complicated net work,all of these were involved in the pathogenesis of psoriasis.
