Objective: Establish the rat model of severe burns.Through thetail vein injection of α-MSH,to observe the effects of α-MSH on inflammatoryfactors and study it’s inhibition mechanism to early inflammatory reaction. Tofurther verify the truth that α-MSH can weaken the inflammatory responseassociated with severe burns.Methods:Clean level of adult healthy SD rats18,male and female unlimited, not in pregnancy and lactation, were randomlydivided into the experimental group1(α-MSH treatment group)、 theexperimental group2(saline treatment group)、the control group (blank controlgroup). Take off the back hair with8%sodium sulfide, and keep theexperimental group of rats’ back in the96℃constant temperature waterfor10s caused20%TBSA Ⅲ degree burn wounds. Rats in experimental group1receive injection of α-MSH solution(1mg/ml) at0h,24h,48h afterburned,the dose is1ml/(kg.d). In the corresponding time,the experimentalgroup2got a injection of physiological saline,the dose is1ml/(kg·d).Thecontrol group normal feeding.Bind up the wound,and change the bandageevery other day.Observe and compare the rats’ wound conditions. At6h,12h,24h,48h,72h after scalded, extraction the venous blood of the18rats for1-2ml to detect blood cytokines IL-6, IL-8levels by ELISA. Finally kill the miceand get the fresh scalded tissue for pathological observation. All of theexperimental data were represented by means±standard deviations (x±s).Differences between groups and within group were compared withsingle factor analysis of variance. Results:(1)、The results of the nakedeye:After scalded0.5h, The rats’ wood were significantly congestion, edema,and with a little effusion, and there is a clear demarcation between thesurrounding tissue.As the time prolonged, the skin becomes pale, glossiness isdim,and with a decreased elasticity. Comparative observation, experimentalgroup1’s black eschar relatively less than experimental group2,and the localinflammation reaction is lighter.(2)、 The expression of inflammatorycytokines:The level of IL-6in the five observation time-points in experimentalgroup1is:(35.2±1.50) pg/ml、(54.08±2.58) pg/ml、(76.23±2.56)pg/ml、(111.56±2.85)pg/ml、(178.64±9.30)pg/ml; The experimental group2is:(39.60±2.16)pg/ml、(59.94±1.78)pg/ml、(88.00±3.29)pg/ml、(122.62±2.80)pg/ml、(190.75±5.27)pg/ml;while the control group is:(31.24±2.33)pg/ml、(32.25±1.10)pg/ml、(31.63±2.83)pg/ml、(30.88±2.64)pg/ml、(31.93±1.80)pg/ml。The level of IL-8in the five observation time-points in experimentalgroup1is:(39.63±1.59)pg/ml、(49.81±1.93)pg/ml、(102.3±7.21)pg/ml、(126.96±7.29)pg/ml、(180.81±5.22)pg/ml;The experimental group2is:(49.98±2.41) pg/ml、(54.10±3.40) pg/ml、(113.07±4.86) pg/ml、(149.95±5.05)pg/ml、(195.51±6.94)pg/ml;while the control group is:(31.52±2.63)pg/ml、(31.86±2.00)pg/ml、(32.74±2.19)pg/ml、(32.60±3.14)pg/ml、(33.98±2.73)pg/ml。The IL-6and IL-8’s expression quantity of the twoexperimental groups showed a trend of increased as time goes by, and alwayshigher than the control group. the IL-6and IL-8expression quantity of experimental group1was lower than experimental group2, the difference wasstatistically significant (P <0.01).α-MSH in reducing inflammatory cytokineson the expression of IL-6and IL-8have statistical significance.The IL-6andIL-8of control group’s expression quantity did not see obvious fluctuations, nostatistical difference.(3)、Histological observation:Use the HE staining analysistwo experimental rats’ terminal wound specimens found there is a wide rangeof inflammatory cell infiltration、swelled and denatured fat cell、expansivefibroblasts、expansive capillaries and part of the capillaries’ tube wall is weak,orembolism necrosis. the degree of degeneration and necrosis In experimentalgroup1is lighter in group2. Conclusion:(1)、α-MSH can reduce inflammatorycytokine IL-6, IL-8levels, inhibit the body’s early systemic inflammatoryresponse after burn, at the same time can reduce the wound local tissue edema,necrosis, swelling and inflammatory reaction.(2)、The role of α-MSH reduceinflammation reaction in the severe burn is obvious proved by the animalexperiment.
