| Objective: Establish the rat model of severe burns.Through thetail vein injection of α-MSH,to observe the effects of α-MSH on inflammatoryfactors and study it’s inhibition mechanism to early inflammatory reaction. Tofurther verify the truth that α-MSH can weaken the inflammatory responseassociated with severe burns.Methods:Clean level of adult healthy SD rats18,male and female unlimited, not in pregnancy and lactation, were randomlydivided into the experimental group1(α-MSH treatment group)〠theexperimental group2(saline treatment group)ã€the control group (blank controlgroup). Take off the back hair with8%sodium sulfide, and keep theexperimental group of rats’ back in the96℃constant temperature waterfor10s caused20%TBSA â…¢ degree burn wounds. Rats in experimental group1receive injection of α-MSH solution(1mg/ml) at0h,24h,48h afterburned,the dose is1ml/(kg.d). In the corresponding time,the experimentalgroup2got a injection of physiological saline,the dose is1ml/(kg·d).Thecontrol group normal feeding.Bind up the wound,and change the bandageevery other day.Observe and compare the rats’ wound conditions. At6h,12h,24h,48h,72h after scalded, extraction the venous blood of the18rats for1-2ml to detect blood cytokines IL-6, IL-8levels by ELISA. Finally kill the miceand get the fresh scalded tissue for pathological observation. All of theexperimental data were represented by means±standard deviations (x±s).Differences between groups and within group were compared withsingle factor analysis of variance. Conclusion:(1)ã€Î±-MSH can reduce inflammatorycytokine IL-6, IL-8levels, inhibit the body’s early systemic inflammatoryresponse after burn, at the same time can reduce the wound local tissue edema,necrosis, swelling and inflammatory reaction.(2)ã€The role of α-MSH reduceinflammation reaction in the severe burn is obvious proved by the animalexperiment. |
