A Study On Self-made Enteritis Enema In Treating Ulcerative Colitis In Active Stage With Damp-heat Symptom Of Rats

Objective: This paper aims at constructing the rat model which is closest todamp-heat human pathogen of ulcerative colitis, observing the self-madeenteritis enema in the treatment of the active phase of an animal model, andevaluating the therapeutic effect of decoction.Methods:1.155healthy3-month-old SD rats, male and female halves,divided into the under groups: blank section (K), model control (M), theChinese medicine treatment group (A1、A2、A3), Western medicine treatmentgroup (B1、B2、B3), saline treated group (C1、C2、C3). On the basis of feedinghigh fat, high sugar and spicy food to rats, the rat model with damp-heatsymptom of ulcerative colitis with the immune complex legal has been made.Group A1、A2、A3employ the self-made inflammatory bowel enema, group B1、B2、B3employ mesalazine enema, and group C1、C2、C3employ saline enema,once a day. the Groups of A1、B1、C1tread1weeks in all，the Groups of A2、B2、C2tread2weeks in all,and the Groups of A3、B3、C3tread3weeks in all.2. Collecting specimens in five times: the first time: after feeding rats2weeks,the group-K were observing the general condition, weighing, testing stool, andextract2ML cardiac blood from K Group rat, doing IL-8and IL-13testing in atest tube. After that, immediately opening the abdominal cavity and extractinganus6cm lesions of colon，and with physiological saline rinse three times,afterrapidly visual score,intercepting obviously pathological bowel about 6cm,putting it in the specimen bottle with paraffin-embedded slice, HE stained,light microscopic observation; the second time: With the successful model,collecting colonic tissue and blood specimen With the above method from ratmodel M-Group; the third time: after treading rats1weeks, collecting colonictissue and blood specimen With the above method from Group of A1、B1and C1;the fourth time: after treading rats2weeks, collecting colonic tissue and bloodspecimen With the above method from Group of A2、B2and C2; the fifth time:after treading rats3weeks, collecting colonic tissue and blood specimen Withthe above method from Group of A3、B3and C3.Compare the indexes,performing statistical analysis. Data were analyzed using mean±standarddeviation (x±s), using the SPSS19.0Statistical software to analyze, more thantwo-sample were compared with single factor variance analysis of variance ofthe comparision test between one and the other. P＜0.05,the difference wasstatistically significant.Results:1. The damp-heat symptom of ulcerative colitis rat model can besuccessfully created through high-fat, high-sugar and spicy die on the basis ofthe the immune complex legal, and the model is stable;2. Use the self-madeenteritis enema to treat rats with damp-heat symptom of ulcerative colitis, rat’ssyndrome significantly improved and disease activity index (DAI) in ratsdecreased significantly (p<0.01);3. Use the self-made enteritis enema to treatrats with damp-heat symptom of ulcerative colitis, pathological biopsy of thecolon showed that colonic mucosa in obvious fix, colonic mucosal damage index (CMDI) significant improved ((p<0.01);4. Use the self-made enteritisenema to treat rats with damp-heat symptom of ulcerative colitis, the immuneindices in serum of rats effects (after modeling, Group M in rat serum IL-8increased significantly compared to Group K (P<0.01), IL-13decreasedsignificantly compared to Group K (P<0.01) and Group of the Chinesemedicine treatment serum IL-8decreased significantly compared to GroupM(P<0.01), IL-13increased significantly compared to Group M(P<0.01);among the there treatment group, serum IL-8in Group of the Chinese medicinetreatment decreased significantly compared to Group of the Western medicinetreatment and saline treatment (p<0.01), and IL-13significantincreased(p<0.05).Conclusion:1. Using high fat and sugar, spicy diet intervention on the basisof immune complex legal can result in a activity syndrome animal model ofulcerative colitis, and the model is stable, Chinese medicine dialecticaltreatment for ulcerative colitis provides a more targeted animal model.2.Theself-made enteritis enema is low cost,can effectively inhibit inflammation,regulate the body’s immune function, improve the blood circulation in intestinalmucosa, promote ulcer healing, relieve systemic symptom, the activitysyndrome ulcerative colitis has good treatment effect, its curative effect isbetter than that of the mesalazine. This experiment is limited to the self-made enteritis enema on the activitysyndrome of damp-heat ulcerative colitis model animal efficacy studies, andclinical application in the treatment of remission remains to be further research.
