Objective: To determine the influence of electro-acupuncture (EA)pretreatment at zusanli (st36) on the content of HPA axis related substances inc-Kit gene knockout rats local skin compare with the wild type rats; and todetermine whether this effect is regulated by mast cells.Methods: Select30albino c-kit knockout rats with an age of6weeks as Agroup;30pigment wild type rats as B group. Those in A and B group weregiven EA penetrating at left zusanli for30min, the right zusanli area skin forblank control. After3months, All rats were killed after EA penetrating, takethe bilateral1cm*1cm skin in zusanli area, each skin was divided into two.Meanwhile, these samples according to the source were divided into fourgroups: gene knockout rats EA side (KE); gene knockout rats none side (KN);wild type rats EA side (WE); wild type rats none side (WN). Respectively,using immunohistochemistry and Western Blot to detect CRH-R1, MC1R,CRH-R2, MC2R, NTR, UBM3.Results:1. Comparing with KN, there was no statistical difference of theconcentration of all6substances in KE (Independent-Sample Test).2. Concentration of all6substances in KN group was higher than WN.However, this difference was not significantly.3. No statistical difference in the concentration of all6substances wasfound between WE and WN (Independent-Sample Test). 4. Comparing the difference of ΔKE (ΔKE=KN-KE) and ΔWE(ΔWE=WN-WE) between the six substances, there was a statisticallydifference of CRH-R1expression in ΔKE comparing to ΔWE (P=0.037); therewas no statistically difference in other5substances between ΔKE and ΔWE.Conclusions:1. Electro-acupuncture has an effect on the content of HPA axis relatedsubstances in local skin;2. Mast cell is very important to the effect of electro-acupuncture;3. Mast cells can induce CRH-R1and have the positive effect in the stress. |
