| Objective:This subject used a model of allogeneic transplant skin to observe whether it could fulfill its immune adjustment function and further improve the skin graft survival rate, to provide support for its clinical application.Methods:1ã€Set Wistar rats for donor, Lewis rats for receptors, take Wistar rats full-thickness skin grafts to Lewis rats back (about2cm x2cm), compression bandage.2ã€According to the different drug,rat random into four groups, control group (control), Tanshinonella group (TANIIa), cyclosporine group (CsA), Tanshinonella+cyclosporine group (TANIIa+CsA).3ã€Skin graft survival, histological appearance were observed and the expression of IFN-γ/TNF-α/IL-2and TGFp1andCD3/CD4/CD8were studied.Results:1ã€Increased skin graft survival was demonstrated in Tanshinonella group compared with the control group and cyclosporine group compared with the Tanshinonella group and Tanshinoneâ…¡a+cyclosporine group compared with CsA group(P<0.05).2ã€Reduced the expression of IFN-yand TNF-αand IL-2was demonstrated in Tanshinoneâ…¡a group compared with the control group and Tanshinonella+cyclosporine group compared with the CsA group(P<0.05).Reduced the expression of IL-2was demonstrated in Tanshinonella group compared with cyclosporine group (P<0.05),but the expression of IFN-γand TNF-α haven’t significant differences between the two group(P>0.05).3ã€Increased the expression of TGFβ1was demonstrated in Tanshinonella group compared with the control group (P<0.05).Expression of TGFβ1of Tanshinonella+cyclosporine group was increased than CsA group(P<0.05).The expression of TGFβ1haven’t significant differences between Tanshinoneâ…¡a group and cyclosporine group (P>0.05).4ã€Reduced the expression of CD3and CD4and CD8in peripheral blood was demo-nstrated in Tanshinoneâ…¡a group compared with the control group (P<0.05).The expre-ssion of CD3and CD4and CD8in peripheral blood of Tanshinoneâ…¡a+cyclosporine group was reduced than CsA group(P<0.05).The expression of CD3andCD4and CD8in peripheral blood havent significant differences betwee Tanshin oneâ…¡a group and c-yclosporine group(P>0.05)Conclusion:Tanshinoneâ…¡a could regulate immunity through increasing the expression of TGFβ1and reducing the expression of IFN-γ/TNF-α/IL-2and CD3/CD4/CD8. Combined medication of Tanshinoneâ…¡a and cyclosporine had better inhibition function for immunity than Tanshinoneâ…¡a alone. |
