| Objective To investigate the influences of BTXA on the number of fibroblasts, collagen fiber density and expression of α-SMA in myofibroblasts after skin grafting, which are suggested they are the factors about the phenomenon of contraction.Methods20skin grafts were randomly divided into Group S (BTXA pre—injection,10skin grafts) and Group D (non-injection, control,10skin grafts). The contraction of skin grafts model were established by excised four full thickness skin(2cmx2cm) in the back of every rabbits with prefabricated template. Observe the wound healing and skin graft situation. HE staining analysis the number of fibroblasts, collagen fiber density. Measure expression of α-SMA with the immunohistochemical staining and computerized image analysis were used to detect the integrated option density of positive staining of α-SMA.ResultsThe wounds healing and skin grafts situations in Group S at post—operation14day had no significant difference compared with Group D. In group S compared with group D, group SB compared with group SZ and group DB compared with group DZ, the inflammatory cells decrease in the dermis, collagen fibers are relatively decrease in density observed by light microscope, α-SMA in Group S were significantly weaker than those in Group D (P<0.01Pa-SMA=0.003) as well as Group SB than those in Group SZ (P<0.01Pa-SMA=0.000) and DB than those in Group DZ(P<0.01Pa-SMA=0.000).Conclusion The effect of BTXA can reduce the number of fibroblasts, collagen fiber density and expression of α-SMA in myof ibroblasts generated in the contraction of process of skin grafts. |
