Polysaccharides was also known as saccharin, widely found in plants, animals,microorganisms, marine organisms and other biological organism, is an importantinformation molecules in addition to protein and nucleic acid in organisms.Polysaccharides with many kinds of biological activity: anticancer, antioxidant,antiaging, antibacterial, immunomodulatory, anticoagulant, hypoglycemic,hypolipidemic, antiallergic, anti-inflammatory and anti-virus. However singlespecies of bioactivity of polysaccharides relatively simple,while the polysaccharidemixture is able to have a variety of biological activities, and synergy between thedifferent polysaccharide polysaccharide mixture has a single polysaccharide can notmatch advantage. Therefore, the study of mixture of polysaccharides research hasimportant theoretical significance and application value.Water extracting and alcohol precipitation from ginseng, matsutake andmushrooms were extracted polysaccharide, respectively, after Sevag deproteinized,depigmentation after oxidation were isolated and purified to obtain purifiedpolysaccharide, preparation, ginseng, Lentinus edodes and mushrooms mixture ofpolysaccharide. Aim to play ginseng polysaccharide, Polysaccharides of TricholomaMatsutake and Lentinan synergistic enhancement of three kinds of polysaccharide andpolysaccharide mixture has a strong biological activity. To CTL cytotoxicity as anindex, by orthogonal experiments showed that the optimal set of squarepolysaccharide mixture of ginseng polysaccharides, Lentinus edodes polysaccharideand Lentinan1:1:1mass ratio.Ginseng polysaccharide, polysaccharides of tricholoma Matsutake, lentinan andmixture of polysaccharide conducted a study on the quality evaluation:(1) The rate of extraction of ginseng polysaccharides, polysaccharides oftricholoma Matsutake and lentinan were8.85%,9.40%and10.5%, respectively.(2) By phenol-sulfuric acid method for the determination of ginsengpolysaccharide, polysaccharide of tricholoma Matsutake, lentinan and mixture ofpolysaccharide of total polysaccharide, the content were62.96%,59.13%,33.86% and53.17%, respectively; using hydroxy biphenyl determination of polysaccharidesuronic acid in ginseng polysaccharide, polysaccharide of tricholoma Matsutake,lentinan and mixture of polysaccharide, the content were16.44%,9.37%,4.47%and9.54%, respectively. Using3,5-dinitrosalicylic acid method measured ginsengpolysaccharide, polysaccharide of tricholoma Matsutake, lentinan and mixture ofpolysaccharide, and did not contain reducing sugar; After pretreatment of the samples,according to the DNS reducing sugar assay method for the determination of ginsengpolysaccharide, polysaccharide of tricholoma Matsutake, lentinan and mixture ofpolysaccharide, the starch content were7.26%,2.8%,3.77%and3.94%,respectively.(3) Liquid chromatography-mass spectrometry determination of monosaccharidecomposition of polysaccharides, ginseng polysaccharide contained three kinds ofmonosaccharide, including glucose, arabinose and rhamnose, the ratio of62.85:3.23:0.79; polysaccharide of tricholoma Matsutake contained three kinds ofmonosaccharide, including glucose, arabinose and xylose, the ratio of115.50:0.20:0.27; lentinan contained five kinds of monosaccharide, including glucose, arabinose,xylose, mannose and fructose, the ratio of3.15:0.16:0.18:3.42:2.27.(4) Efficient use of gel permeation chromatography and molecular weightdistribution of the polysaccharide, ginseng polysaccharides contained a numberaverage molecular weight polysaccharide2.08009×106,5.51902×105,1.43566×105,1.8667×104,3.386×103and5.48×102, polysaccharide of tricholoma Matsutakecontained a number average molecular weight of2.58932×106,1.38786×105,3.6868×104,1.1588×104,4.83×102, and6.3×10; lentinan contained a number averagemolecular weight of1.88721×106,3.21322×105,1.09038×105,2.2555×104, and5.22×102.Ginseng polysaccharide, polysaccharide of tricholoma Matsutake, lentinan andmixture of polysaccharide for stability research, at high temperature conditions, totalpolysaccharide content declined not over7%; in strong light conditions, totalpolysaccharide content declined over10%, indicating that polysaccharide shouldstored in the dark; high humidity conditions, moisture weight gain of more than14%,indicating that polysaccharide is easy to absorb moisture, drying should be confinedpreservation. After addition of different pH solution, in the oxidant (H2O2) andreducing agent (Na2SO3) conditions, polysaccharide quality as conditions change, the absorbance value is not obvious, indicated that polysaccharides have good pHstability and resistance to oxidation and reduction.Ginseng polysaccharides, polysaccharides of tricholoma Matsutake, lentinan andmixture of polysaccharide for hygienic detection studies, ginseng polysaccharide,polysaccharide of tricholoma Matsutake, lentinan and mixture of polysaccharide ofbacterial were120,0,0and50CFU/g,respectively, Ginseng polysaccharides,polysaccharides of tricholoma Matsutake, lentinan and mixture of polysaccharidewere not controlled detection of E. coli bacteria, Salmonella, Shigella, Staphylococcusaureus, in line with the2010edition of the Pharmacopoeia of China microbial limitstandards..
