As a Chinese traditional medicine raw material, deer fetus has been used forhundreds of years, for its abundant nutritious function. As it contains rich proteins,peptides, amino acids, nucleic acids, vitamins and plenty of hormones and enzymes, ithas great potential in regulating menstrual function and beautifying function.However, the manufacturing technique is remarkably undeveloped, further study isdesperately needed.In this work, domesticated deer was used as the raw material to fabricategranules. After grating in the colloid mill with6times water for8mins, the deer fetuswas hydrolyzed with trypsin. We used response surface methodology (RSM) toachieve the optimumenzymolysis condition:2028U/g trypsinat45.4℃, PH7.5for3.12h. Then the residue of enzymolysis was extracted in75%ethanol for48h at roomtemprature. The final extract of deer fetus was constituted of the alcohol extract andtrypsin hydrolysate.Adding adjuvantproportionally into deer fetus extract (60%deerfetus,20%brown sugar,5%jujube,5%medlar,5%soy isoflavones,5%honey, allthese adjuvant materials were boiled for2h together to get the supernatant as finaladjuvant). After drying by spray dyer, the deer fetus extracts powder we got was usedto fabricate granules. We used orthogonal test to draw the optimum granuesfabrication condition: the ratio of accessoriesis extraction: accessories=1:1, thecompositionof accessories is soluble starch: sucrose powder=1:0.25, concentration ofhumectantethanol is75%, drying tempratureis60℃.In this thesis, we well established a preliminary quality control standard for ourproduct. The finaldeer fetus granules are brown, sweet smelling and tasting, withoutvisible impurity. The product has homogeneousappearance, as well as excellentstability. Our produt also reaches the related standards of Chinese pharmacopoeia ondifferent tests, such as moisture content test, ash content test, and microbial limit test.Experimental data shows that our granules of fetus from domesticated deercansignificantly upregulate the spleen and thymus index of mice. The resulted spleen and thymus index, as well as the positive result of MTT test indicates that our productsplay a crucial role in the enhancement of immunity. To sum up, with positive effect inupregulating the immunity of mice, deer fetus has promising future as health product.
