Application of deer products in China has a long history. Velvet and placenta arenot only traditional Chinese herbal in our country but also extremely valuable tonic.In addition to velvet, other parts of deer can be used as medicine, but in velvet andplacenta manufacturing process there are a lot of unused byproducts, such as heart,liver, spleen, venison, etc of deer. there are very few people to research on theseproducts, but they also have a high commercial value.In this paper, spleen of domesticated deer contains a variety of biologicallyactive ingredients, including high protein, small peptides, polysaccharides, ribosecontent. In this paper, we saw spleen of domesticated deer as raw material, establisheda reliable measurement method by Bradford to determine concentrations of Proteinand peptides to calculate the extraction rate. We draw the optimum: water feed ratio of12:1, cutter grinding time7min, Papain1500U/g,43℃, shaking5.3h, and thencentrifugat5920×g15min, by using response surface methodology in protein,peptide extraction rate index, optimizating extraction conditions and hydrolysisconditions. The extract is the supernatant liquid. At this time peptide extraction rate ismore than103%before optimization. Then we used extraction to prepared granules.We draw the optimum, extraction: accessories=1:1, soluble starch: sucrosepowder=1:0.25, concentration of ethanol was75%, the granules were dried at60℃,by using Orthogonal experiment in particles passing rate, moisture absorption rateand angle of repose, optimizating preparation of formulations.We builded quality standards of the Granules of spleen from domesticated deer.This product is a tan to brown uniform particles, gas micro, sweet, no visible foreignmatter, uniform size, good melt. Moisture content is no more than5%. The total ashcontent is not exceeding0.7%. Peptide content is no less than36mg/g. Microbial limitis in line with national standards. The preparation has excellent stability bypreliminary stability study. This paper studied immunocompetent of the granules of spleen fromdomesticated deer according to the provisions of Health food function evaluationprocedures and test methods. In Determining NK cell killing active,we know whenthe target ratio was1000, active were all above50%. Medium and high dose groupsof mice was significantly higher than the negative control group. Low dose groupactivity of NK cells was quite positive control. Lymphocyte transformation test,delayed-type hypersensitivity were positive. Serum hemolytic of experimental groupwere also increased.The granules of spleen from domesticated deer we developed has stable quality,clear immune function and a market prospects. This study Provides a new way ofthinking in development of industrial by-products of domesticated deer and alsoprovides the basis for the theory into actual production.
