Objective:Through the establishment of the anteriordisc displacement (AD) animal model, evaluate the influence of different joint loading on condylar cartilage to the expression of IL-1β. Explore the relationship between different pressure and pressure time and the expression of IL-1β by the separated chondrocytes was cultured in vitro, then study the correlation of chondrocytes IL-1β expression differences and temporomandibular joint under different stress. Thus it will provide the basis for elucidating the molecular mechanism of the function of IL-1β in temporomandibular joint cartilage.Methods:In this study, the rabbit AD model was established by surgical procedures, and all AD rabbits were sacrificed at different times after surgery, then their cartilage tissue was achieved for immumohistochemical staining and the measured of IL-1βpexpression. In addition, the separation condylar cartilage cells were cultured in vitro. Then detected the cartilage cells morphology and growth states by immunocytochemistry and MTT assay. The different static pressure environment in vitro including the pressure size and loading time was simulated in the culture process and evaluated the IL-1β gene expression for statistical analysis by cellular RNA extraction, real-time quantitative reverse transcription-polymerase chain reaction (RT-PCR) and Western Blot.Results:In AD model, the results showed that the expression of IL-1β in condylar chondrocytes hypertrophy layer was significantly higher than others (P<0.05). It showed that the morphology and density of the condylar chondrocytes were significantly different from the other chondrocytes by immunohistochemical staining and MTT assay. By simulating different static pressure environment in vitro, we found the expression of IL-lβwas significantly correlated with joint load level and pressure time, the difference was statistically significant (P<0.05).Conclusion:IL-iβ is so important in the growth and development of condylar cartilage, and the expression of IL-1β was significantly affected by the increase of joint load level and pressure time, thereby affecting the build of condylar organization and function of condyle condylar cartilage cell.
