The Inhibitory Action Of Heating Clearing Therapy On Human Leukemia Cell Line And Primary Cell Collected From AML Patients

[Objective]To observe the inhibitorty effect of heating clearing therapy on human leukemia k562,U937,HL-60cells and the mononuclear cells(MNC) collected from acute myeloid leukemia(AML) patients. Investigate the significance of the micro information for TCM syndrome and provide the basis for clinical diagnosis of TCM,also provide the reference and support for the standardization of combining traditional Chinese and western meidicine treatment.[Methods]PART1:To selected SD rats,prepared the herb serume containing xijiaodihuang tang(xijiao),Ara-c and DNR(DA),combining xijiao and DA,normal saline with serolojic pharmaeological method and treated the leukemia k562,U937,HL-60cells last for24,48,72hours with herb serum in5%,10%,15%concentration.The effects of the herb serum the proliferation of k562,U937,H1-60cells are observed with morphology and with MTT assay.PART2:To selected AML patients comed from Fujian medical university affiliated Union hospital,collect patients’four diagnostic information,select heat syndromes with differentiation of symptoms and signs,take the MNC form the patients’ bone marrow,and treat MNC last for24,48,72hours with herb serum in5%,10%,15%concentration. The effects of the herb serum the proliferation of MNC is observed with morphology and with MTT assay.[Results]PART1:1.The herb serum containing xijiao,DA,xijiao+DA in5%,10%,15%concentration could influence the morphology of leukemia k562,U937,HL-60cells.The amount of the cells treated by the different concentration is less.and the state growth is poor.the cell form is irregular,contraction and narrowed,and appear cell fragment,especially by the group of xijiao+DA. 2. The four group herb serum in5%,10%,15%concentration could inhibit the proliferation of k562,U937,H1-60cells,especially HL-60cells.The inhibition to K562.H1-60after treatment with xijiao group was no significant differences between different concentrations,but the inhibition to U937was significantly increased in a dose-dependent manner. The inhibition to K562,U937after treatment with xijiao+DA group was no significant differences between different concentrations,but the inhibition to HL-60was significantly increased in a dose-dependent manner.PART2:l.The herb serum containing xijiao,DA,xijiao+DA in5%,10%,15%concentration could influence the morphology of MNC collected from12AML patients.The amount of the cells treated by the different concentration is less,and the state growth is poor,the cell form is irregular,contraction and narrowed,and appear cell fragment,especially by the group of xijiao+DA.2. The herb serum containing xijiao,DA,xijiao+DA could inhibit the proliferation of k562,U937,H1-60cells,especially HL-60cells,and the inhibition have obvious difference.2.The inhibition to leukemia cells after treatment xijiao+DA group herb serum was higher than xijiao,DA group.3.The heating clearing therapy has curative effect on AML,and the method of differentiation of symptoms and signs could provided basis for AML clinical TCM treatment.
