Objective:To explore the effect of Huangqijiedu Decoctionon apoptosis of MCF-7and expression of Notchl, Jagl and CyclinD of human breast cancer cells in vitro and to reveal its mechanism of inhibiting recurrence and metastasis of breast cancer.Methods:First of all to prepare drug-containing serum,12female SD rats were randomly divided into four groups:Huangqijiedu decoction serum group, Doxorubicin serum group,Shenyijiaonang serum group and normal saline serum group. Different groups’rats were given by gavage two tims a day for3days with the same concentration of Huangqijiedu solution, Doxorubicin solution, Shenyijiaonang solution and normal saline solution.In fourth days after seventh gavage, narcotize rats with chloral hydrate, drawing blood from the abdominal aorta.After the intervention of different drug-containing serum on MCF-7,using MTT inspects its effect on cell inhibition,Flow cytometry inspects its impact on apoptosis of MCF-7breast cancer cells in vitro experiment,RT-PCR inspects its influence on Notchl and Jagl genes,Western-blot inspects the effect on CyclinD1expression.Results:1.MTT:compared with the normal saline serum group,all the drug-containing serum groups have the function of inducing the apoptosis of MCF-7breast cancer cells.In addition to the5%concentration group,the OD ratio between different drug-containing serum groups and normal saline serum group have statistically significant difference(P<0.05), and the highest inhibition rate is Doxorubicin serum group, followed by Huangqijiedu decoction serum group, at last is Shenyijiaonang serum group; 2. Flow cytometry:all the drug-containing serum groups have the function of inducing the apoptosis of MCF-7breast cancer cells.the apoptosis rate between different drug-containing serum groups and normal saline serum group have statistically significant difference(P<0.05), and the highest apoptosis rate is Doxorubicin serum group, followed by Huangqijiedu decoction serum group, at last is Shenyijiaonang serum group;3. RT-PCR:compared with the normal saline serum group,all the drug-containing serum groups can inhibit the expression of Notchl, Jag1of MCF-7.The inhibition rate between different drug-containing serum groups and normal saline serum group have statistically significant difference(P<0.05), and the highest inhibition rate is Doxorubicin serum group, followed by Huangqijiedu decoction serum group, at last is Shenyijiaonang serum group;4.
