This paper mainLy studies the extraction process of resveratroL from PoLygonum cuspidatum and macroporous resin for purification of resveratroL. It uses uLtravioLet spectrophotometry and high performance Liquid chromatography to track and monitor。ResveratroL is naturaL anti-oxidants, with Lower bLood viscosity, inhibiting pLateLet aggreagation and vasodiLation to maintain bLood fLow and other functions. ResveratroL can prevent cancer occurrence and deveLopment. It can aLso resist aLsoanti-atheroscLerosis and coronary heart disease, high choLesteroL prevention roLe. It is increasingLy wide range of appLications. At present resveratroL is mainLy extracted in PoLygonum cuspidatum. Diverse separation methods are increasing. The big empty adsorption resin to separate purified resveratroL from PoLygonum cuspidatum method aLso is not mature enough. Moreover, With the increase of resin types, expLoring the optimum extraction conditions and researching for different resins on the separation and purification of resveratroL in PoLygonum cuspidatum have an important significance.This paper conducted a triaL of resveratroL organic soLvent extraction. It examined the impact of various factors affecting the extraction of resveratroL. According to the singLe-factor test and the orthogonaL test, the optimaL extraction conditions were determined. They are voLume fraction of ethanoL,50%, water bath extraction temperature 80℃, the soLid-Liquid ratio of 1:100. Taking into account the temperature is too high, ethanoL, voLatiLe faster, so the actuaL extractions take:ethanoL voLume fraction of 60%. Extraction temperature is 60℃, the soLid-Liquid ratio of 2:100. ResveratroL an extraction rate is 82.37%.This paper aLso examined the LS-303 resin and LD-601 macroporous resin for purification of resveratroL. To uLtravioLet-visibLe spectrophotometry and high performance Liquid chromatography before and after purification of resveratroL in PoLygonum cuspidatum extract and its impurities changes. Experiments incLuded a static test and dynamic test of two parts。Study of these two kinds of resin adsorption amount of resveratroL, adsorption kinetics, adsorption isotherm adsorption, etc., as weLL as pH, concentration, fLow rate and other factors on the adsorption performance. The resuLts show that:ResveratroL in acid conditions and 70℃beLow has a good stabiLity; Static adsorption equiLibrium time is about 60min, adsorption process in Line with constant equation of ELovich equation and doubLe-constant equation. pH at 2 to 8, within, LS-303 resin and LD-601 resin has a good absorption of resveratroL, and the adsorption process is spontaneous and exothermic process of thermodynamics. When the coLumn Length 5.0cm, diameter 1.0cm, buiLt-3.0000g When the coLumn Length 5cm, diameter 1.0cm, buiLt-3.0000g resin, when the fLow rate is 1.0mL/min, concentration 20μg/mL, LS-303 resin and the LD-601 resin on the adsorption rate of resveratroL, respectiveLy 99% and 94%. ResveratroL has been adsorbed with 60% ethanoL soLution may be a better eLution. Experiment tracking and monitoring by HPLC. LS-303 resin and LD-601 resin have a better roLe in the purification of resveratroL. The resin content of a purified is 38.71%, respectiveLy, and 26.85%.
