Objective:Based on the previous studies of Jing-fang powder, this study investigate the effective parts of anti-allergic effect of Jing-fang powder and its mechanism, by using allergy mouse model as PCA and other experimental models. So further provide the pharmacological basis for the compatibility of Jingjie and Fangfeng to cure diseases caused by "wind cold" and interpret the modern scientific connotation.Methods:(1) Extract different polarity parts of Jing-fang powder and screen the effective parts of anti-allergic effects:①System solvent method, use petroleum, ethyl acetate and N-butanol sequentially extract the decoction, get petroleum constituent, ethyl acetate constituent, n-butanol constituent and Water Extraction constituent.②Use DNCB-induced delayed-type hypersensitivity of mice,screening the efficient extraction site of anti-allergic effects from Jing-fang powder.Adopt histamine,5-HT induced increase in capillary permeability experiments in mice, dextran-induced pruritus in mice and the homologous/heterogeneous PCA experiments to verify the anti-allergic effects of effective extract.(2) Search the mechanism of anti-allergy effects of the effective extraction from Jing-fang powder:①Measure the serum IgE and LTB4levels, measure the lung tissue NO, iNOS, LTB4levels of initiative systemic allergic model mice by ELISA.②Measure the lung tissue PGE2levels of model mice by UV spectrophotometric determination.③Measure the IL-4, IFN-ymRNA expression levels of initiative systemic allergic model rat by RT-PCR.④Measure the serum IL-4, IFN-y levels of model rat by ELISA.Results:(1) The volatile oil, decoction with volatile oil and ethyl acetate extract can significant inhibit DNCB-induced ear swelling and ear vessels permeability of delayed hypersensitivity model mice (P<0.05),and other groups are not obvious.(2) Continuous gavage ethyl acetate extract from Jing-fang powder6.67g/kg,10.01g/kg for nine days, significantly reduced DNCB-induced mouse ear edema of delayed hypersensitivity model mice,and reduce the thymus, spleen index (P<0.05, P<0.01); significantly inhibit the histamine and5-HT-induced mouse skin capillary permeability (P<0.05). Continuous gavage ethyl acetate extract from Jing-fang powder10.01g/kg for seven days, can significantly shorter dextran-induced pruritus duration in mice (P<0.05).Continuous gavage6.67g/kg,10.01g/kg for six days, can significantly reduce the abdomen skin capillary permeability increased of homologous/heterogeneous PCA model mice (P<0.05, P<0.01).(3) Ethyl acetate extract of Jing-fang powder6.67g/kg can significantly lower ova-induced allergic mouse thymus index and spleen index (P<0.05).significantly lower serum IgE, LTB4levels (P<0.05), and significantly reduced lung tissue NO, LTB4and PGE2levels (P<0.05). Ethyl acetate extract10.01g/kg,6.67g/kg can significantly reduce lung tissue iNOS levels (P<0.05).(4)Ethyl acetate extract of Jing-fang powder2.5g/kg,5g/kg significantly lower the expression of IL-4mRNA from tissue of active systemic allergy model rat and lower the serum IL-4levels. At the same time, they can significantly increase the expression of IFN-ymRNA from tissue of and elevated serum IFN-γ levels. To rectify the imbalance of Thl/Th2cell subsets function.Conclusion:The ethyl acetate extract is the efficient extraction site of anti-allergic effects from Jing-fang powder. Anti-allergic mechanism is related to reduce the formation of IgE, inhibit the release of inflammatory allergic mediator NO, LTB4and PGE2, that is meaning the ethyl acetate extract intervene the arachidonic acid metabolism pathway and NO/iNOS metabolic pathway. In addition, the drug lower the expression of IL-4mRNA and increase the expression of IFN-γ mRNA of model rat. It can also lower serum IL-4levels and elevate serum IFN-γ levels.To exert anti-allergic effects by correcting the imbalance of Thl/Th2cell subsets function.
