Objective:Copy rats model on chronic bronchitis, giving xiao cheng qi decoction to treatment groups, at the same time. Choosing time points to observation lung and bowel tissues expression level of transforming growth factor (TGF-beta1), the receptor (TGF-beta R I) and the epidermal growth factor (EGF) and the receptor (ErbB3) expression level of model groups and treatment groups. By this way, herbs treatment mechanism on "treating intestinal diseases to relieve pulmonary diseases" would be explored.Methods:Select60male SD rats and randomly divided into treatment groups (20), blank model group (30) and groups (30) according to weight. Put the blank group rats into smoke-free environment, meanwhile, put the model group rats into the smoke environment in order to establish pulmonary disease (chronic bronchitis) animal model, totally smoked70days. The treatment group rats choose small gas bearing decoction treatment, in the building after20days,1times a day. Treatment group randomly drew10,50d and70d after building to be put to death. Observe pathological morphology change of lung, stomach, duodenum, jejunum, ileum, colon, and rectum. Test TGF-β1、TGF-βR1、Smad3、EGF、ErbB3expressions in lung and colon tissue of rats.Result:1. Pathological morphologyLight:Building after20days, the lung tissue anatomy model group10rats in bronchial epithelial cells widely degeneration, necrosis, fall off, partial bronchial mucosa associated with inflammatory cells infiltration. Local hyperemia and edema, two rats colon tissue lesions of intestinal wall epithelium cell degeneration, necrosis, inflammatory cells infiltration, a small number of colon mucosa of rats was widely erosion, now has "into the intestine pathological changes in lung tissue. Building after50days, anatomical model group10rats lung tissue pathological morphological changes are visible to the lung tissue structure, bronchial epithelial cells visible widely degeneration, necrosis, fall off, with a large number of inflammatory cells infiltration, a small number of rat bronchial lumen with inflammatory exudation, part with layer appear inflammatory cells infiltration, seepage, pulmonary interstitial cells arranged disorder, fibrosis change, accompanied by a small amount of fiber cell damage.70%of colon of rats structure of the organization of pathology changes obviously, lens more than can be found under the stove sex erosion venereal stove, located in the colon tissue superficial, visible epithelial cell degeneration, necrosis, fall off, with inflammatory cells infiltration. Glands structure is irregular, mucosal and tissue inflammatory cell infiltration;70days, after the building model group10autopsy pathological morphology change in rat lung tissue, bronchial epithelial cells visible widely degeneration, necrosis, fall off, accompanied by a large number of inflammatory cells infiltration, can be seen in the pulmonary interstitial inflammatory cell infiltration.40%rats pulmonary interstitial structure disorder, fibrosis change, visible repair cells in airway inflammatory exudation cavity is part of the fiber.100%rats colon tissue pathological changes all appeared, intestinal mucosa of multiple stove STD principle and oven with crypt abscesses, partly visible focal epithelial cell degeneration, necrosis, fall off, and the structure of irregular glands, visible mucous membrane and infiltration of inflammatory cells and fibrous structure disorder within organizations. The bronchial epithelial cells of lung tissues were wide degeneration, necrosis and different degree inflammatory cells in model group rats on the20th,50th and70th days. There are respectively20%、70%、90%of the colon tissues appeared local congestion edema and the epithelial cells were wide degeneration and necrosis in model group rats on the20th,50th and70th days. The colon tissues of some rats appeared mucosal surfaces were incomplete and the glands arrangement weren’t regular. Mucosa and sub mucosal cells were wide inflammatory. The duodenum, the jejunum, ileum, rectum, stomach were all normal.2. TGF-β1、TGF-βR1、Smad3、EGF、ErbB3expressionCompared with blank control group, the expression of TGF-β1、TGF-βRI、 Smad3、EGF、ErbB3increased significantly in lung tissue and the expression of TGF-β1、TGF-βR1、Smad3、EGF、ErbB3increased significantly in colon tissue in model group rats on the20th,50th and70th day (P<0.05or P<0.01). Compared with the20th day, the expression of TGF-βR1、Smad3increased in colon tissue in model group rats on the50th days (P<0.05or P<0.01), the expression of TGF-β1、TGF-βR I、smad3、ErbB3decreased in lung tissue and TGF-βR I、smad3、ErbB3in colon tissue of model group rats on the70th days(P<0.05or P<0.01). Compared with the50th day, the expression of TGF-β1、EGF、ErbB3decreased in lung tissue and the expression of smad3decreased in colon tissue in model group rats on the70th days (P<0.05orP<0.01).Conclusion:1. It was a sustained chronic process of pulmonary disease involving large intestine.It took about50d for model rats’chronic bronchitis causing large intestine injury.2. TGF-β1may be one of the material basis about pulmonary disease affected large intestine.TGF-β1/Smad3signaling pathway may participate in the pathologic transmission.3. By giving small bearing gas soup treatment group rats after treatment, based on observation found that, in the50days of lung and intestinal tissue around the TGF-P, smad3and EGF expression decreased obviously, suggests that effective treatment.4. Dredging intestinal organs is the main function of xiao cheng qi decoction. Treatment group rats lung tissue TGF-P, smad3and EGF inflammatory mediator expression decline occurred mainly in lung and intestine pathological change the required point in time, show the improvement of the treatment group rats lung lesions may be related to the improvement and intestinal lesions.5. Herbs treatment mechanism of xiao cheng qi decoction on "treating intestinal diseases to relieve pulmonary diseases" intervention mechanism may related to lower lung, intestinal tissue TGF-p, smad3and EGF expression of inflammatory mediators.
