Study On Mechanism Of Guizhishaoyaozhimu Decoction In The Treatment Of Gout

Objective:Through the detection of acute gouty arthritis model rats andhyperuricemia model rats before and after use of Guizhishaoyaozhimu Decoctionankle volume change PGE2, LTB4and5-HT and the content of rat,Joint soft tissuechanges of TGF-β1, TGF-β R1expression and serum urea nitrogen (BUN),creatinine (Cr) and uric acid (UA) content changes of indicators to explore themechanism of Guizhishaoyaozhimu Decoction in the treatment of gout.Methods:The uric acid sodium injection in articular cavity preparation systemmodel in rats with acute gouty arthritis, ankle swelling volume determination method;EILSA method for the determination of serum PGE2and LTB4content; fluorescencedetection of5-HT content by spectrophotometry; immunohistochemistry joint softtissue transforming growth factor (TGF-β1, TGF-β R1) expression.The suspensionof adenine and ethambutol hydrochloride (adenine100mg·kg–1, ethambutolhydrochloride250mg·kg–1miscible in distilled water) andvolume(1mL·100g–1)stomach perfusion in rats with hyperuricemia animal model,using edge administration side molding method, continuous administration for21days. Respectively at0d,7d,14d,21d blood testing rat blood urea nitrogen (BUN),creatinine (Cr) and uric acid (UA) concentration.Result:Traditional Chinese medicine for acute gouty arthritis model rats with high, middle dose group significantly reduced ankle swelling, there were significantdifferences compared with the model group(P<0.05).Compared with the model group,Chinese medicine of high, low dose group were significantly decreased in the serumcontent of PGE2(P<0.01), no significant difference compared with the positivecontrol group(P>0.05).Compared with the model group, Chinese medicine in highdose of LTB4group were obviously decreased(P<0.01).Significantly decreasedcompared with the model group, Chinese medicine in the synovium of high, low dosegroup5-HT content(P<0.05),the high, middle dose group compared with the modelgroup, there was significant difference(P<0.01).Expression levels compared with themodel group, low dose group, high in rat articular soft tissue of TGF-β1, TGF-βR1significantly enhanced(P<0.05).Hyperuricemia model rats serum BUN, Cr, UAconcentration,7d,14d always compared with the model group, low dose group, theserum levels of BUN, Cr, UA were obviously decreased(P<0.05),21d when comparedwith the model group, high dose group, in BUN, Cr, UA content decreasedsignificantly(P<0.01).Conclusion:After5days of treatment with Guizhishaoyaozhimu decoction caneffectively inhibit the acute gouty arthritis rat articular swelling,Guizhishaoyaozhimudecoction can significantly reduce the serum concentrations of PGE2, LTB4,decreased peripheral5-HT content, and promote joint rat model of soft tissue of TGF-β1, TGF-βR1expression, so as to reduce inflammation, easepain.Guizhishaoyaozhimu decoction can reduce hyperuricemia rats serum uric acidUA, Cr content of serum creatinine and serum urea nitrogen BUN.
