Ojective:Lomatogonium rotatum, the traditional mongolian medicine, was dried herbs of the family Gentianaceae. In this study, we have investigated the chemical composition of Lomatogonium rotatum using stratography and spectroscopy methods.Methods:The5.0kg of dried herbs fine powder of Lomatogonium rotatum was extracted three times with95%alcohol and the extractum was suspended with water. Successively, the suspended extractum was extracted with the same amount of petroleum ether, chloroform, acetoacetate and n-butanol one by one. Then four kinds of various extracted composition were eventually obtained. The monomer compounds were isolated from the extracts extracted with chloroform and acetoacetate by means of silica gel column chromatography, hydroxypropyl sephadex column chromatography, macroporous resin approach, half-preparation thin-layer chromatography and high performance liquid chromatography respectively and in turn. Finally, several spectroscopy methods, including1H-NMR,13C-NMR,2D-NMR (HSQC、HMBC), were applied to identify the structures of these monomer compounds.Results:Eight monomer compounds were, finally obtained and identified in all. There were six monomer compounds extracted with chloroform, including erythrocentaurin,1,8-dihydroxy-4,5-dimethoxy-6,7-methylenedioxyxanthone,1-hydroxy-3,7,8-trimethoxyxanthone,1,8-dihydroxy-3,4,5-trimethoxyxanthone,8-dihydroxy-1,3,5-trimethoxyxanthone and1-dihydroxy-3,5,8-trimethoxyxanthone. There were two monomer compounds extracted with acetoacetate, including1,4,8-trimethoxyxanthone-6-O-β-glucoronyl-(1'6)O-β-D-glucoside and1,2,6-trihydroxyxanthone-8-O-β-D-glucoside.Conclusions:There monomer compounds,1,8-dihydroxy-4,5-dimethoxy-6,7-methylenedioxyxanthone,1,4,8-trimethoxyxanthone-6-O-β-D-glucoronyl-(1'6)O-β-D-glucoside and1,2,6-trihydroxyxanthone-8-O-β-D-glucoside, were new ones first isolated. The present study was a valuable reference for illuminating the pharmacodynamic fundament of Lomatogonium rotatum, formulating the quality criteria and developing new drugs.
