Research On Colloidal Gold Strip Of Chloramphenicol

Chloramphenicol is belongs to broad-spectrum antibiotic has been used in animal husbandry andaquaculture widely. Due to its strong toxic and side effects, chloramphenicol has been forbid by FDA,EU and China. Because of its stable antibacterial property and low price, the phenomenon of illegallyadded is still common. Colloidal gold immunochromatography technique is based on the specificitycombined of antigens and antibodies, aimed to enlarge the effect of reaction and get the result of testingwith human eye.The main purpose is to get study on the preparation and quality of colloidal gold strip andconfirmed by UPLC-MS/MS.1. The colloidal gold with20nm was prepared with a reduction of chloroauric acid by sodiumcitrate. The immunogold was prepared with PcAB, which the concentration was300μg/mL.2. The manufacturing process of colloidal gold test strip of chloramphenicol was consummated.The nitrocellulose membrane which is135s was production of Millipore, and the whole blood filtermembrane which was treated with0.5%tween water solution was used as sample pad.3. The sensitivity for detection of chloramphenicol was1ng/mL,5ng/g,3ng/g by eye in raw milk,honey and chicken eggs. The sensitivity of the test strip was confirmed by UPLC-MS/MS as well. Thegoal of testing different kinds of samples by one kind of test strip has been achieved.
