Chloramphenicol(CAP) is an effective broad-spectrum antibiotics commonly which used in the treatment of human and animal bacterial diseases. But it has adverse effect against human bodies.Therefor,it is important that detct Chloramphenicol in food of animal origin.There are many high sensitivity, good accuracy of detection methods analysis maths for chloramphenicol in the traditional.But it is difficult to meet the on-site, rapid detection needs. A rapid, efficient detection methods and the corresponding product prepared is the trend.In this study, to improve the sensitivity and stability of colloidal gold strip, different size colloidal gold test strip and sample preparation method is optimized.the specific contents are as follows:(1) Optimization of colloidal gold strip craftAccording to the principle of immunochromatography and preparation of chloramphenicol colloidal gold test strip. Colloidal gold particles was prepared by sodium citrate reduction, size of which was 20nm,30nm and 40nm respectively. Each size of the colloidal gold particles were used to prepare the colloidal gold Chloramphenicol test strips and the technical condition of these strips were optimized. The three colloidal gold labeled monoclonal antibody can use the same way, but there is slightly different in protein marker. Three gold standard antibody purification centrifugation speed were 10000,8000,8000 r/min respectively.The smaller particle size the greater the centrifugal speed.Three colloidal gold particle test strip can be shared the same material,just in the nitrocellulose membrane antigen, sheep anti-mouse IgG concentration of the best package is slightly different.(2) Performance Test strips20,30,40nm colloidal gold test strip sensitivity is 7,3,9 ng/mL respectively. And penicillin, gentamicin, streptomycin, tetracycline cross-reactivity were more than 200ng/mL.It is can save 2 months in dry. the 30nm colloidal gold test strip was more stable and sensitive than the others. These results illustrated that the 30nm colloidal gold is more suitable as the marker for chloramphenicol monoclonal antibodies. (3) The actual sample testingUse 30nm colloidal gold production Chloramphenicol test strip,and actual samples will be tested.Strip test results stable and reliable. Compare with traditional methods, solid phase extraction technology not only can be better extracted from the sample target, and achieve the purpose of enrichment.From another angle to improve detection sensitivity.
