Objective:To investigate the relationship of ORMDL3gene polymorphism and childhood asthma in NingboMethods:The subjects included two groups:(1) children with asthma group:From October2011to October2012,150patients with asthma were collected including82male cases and68female cases, with an average age of6.6±1.7years.(2) control group:a total of150healthy children were collected, including80male cases and70female cases, with an average age of7.5±1.3years. DNA extraction kit was applied to extract genome from the experimental subjects. Polymerase chain reaction technique and DNA sequencing methods were used to determine the ORMDL3gene polymorphisms in150cases of childhood asthma and150cases in normal control group. ORMDL3gene rs7216389, rs4378650and rs11650680allele frequencies, genotype and allele frequencies in the two groups were statistically analyzed and compared.Result:(1) There were no statistical difference in sex and age in two groups including the children with asthma and normal control group.(2) There were three genotypes of ORMDL3gene rs7216389, namely TT, TC and CC, in asthma group and normal control group.ORMDL3gene rs7216389locus genotype frequencies in children with asthma and normal control group was statistically significant (P<0.05). Further analysis revealed that,ORMDL3gene rs7216389locus C and T alleles were significantly different (P<0.05).(3) ORMDL3gene rs4378650locus has three genotypes, i.e. TT, TC and CC in the two groups.ORMDL3gene locus genotype frequencies of rs4378650was significantly different between the patient and control groups(P<0.05). And the frequency distribution of C and T was significantly different (P<0.05).(4)ORMDL3gene rs11650680from children with asthma and normal control group had TT, TC and CC genotypes. ORMDL3gene rs11650680genotype frequency distribution in asthma group was significantly different from the control group (P<0.05). In addition, rs11650680locus C and T alleles were significantly different in these two groups (P<0.05).Conclusion:(1) This study found that ORMDL3gene rs7216389, rs4378650and rs11650680sites with single nucleotide polymorphisms (SNPs);(2) There were significant differences in frequencies of allele and genotype in ORMDL3rs7216389, rs4378650 and rs11650680polymorphisms between patients and controls (P<0.05);(3) Our study demonstrated that the ORMDL3rs7216389, rs4378650and rs11650680polymorphisms were associated with the childhood asthma in Ningbo.
