Cervical cancer is one of the leading cause of death of patients with gynecological cancers. Its incidence is rising slowly.In our study, the Hela cells were treated with various concentrations of propolis, and investigated the effects of propolis on growth activity and cisplatin resistance of the Hela cells, and provided theoretical guidance for the clinical application of cancer inhibitors.Methods1. IHC assay was used to determine the influence of propolis on the cytosine secretion in Hela cells.2. The effects of propolis on proinflammatory-related factors were investigated in DNA transcription level, RNA level and protein level.3. The effects of propolis on the Egrl, p21and Ki67expression in the Hela cells were examined using RT-PCR and Western blot.Results1. The secretion of IL-8, TNF-a and IL-10were increased when propolis at a concentration of4,8,16,25μg/mL added to the cell cultures. Meanwhile, the level of NO in macrophage reached seven folds castrated to the blank when was treated with propolis of 16μg/mL.2. The expressions of iNOS、PTGS-2and IL-8in Hela cells can be inhibited by propolis at a broad level DNA transcription level, RNA level and protein level. The results have demonstrated that NF-κB cell signal pathway related proteins played a very important role in these cases.3. The expression level of Ki67was obviously decreased, otherwise, the level of Egrl and P21were both upregulated, when the Hela cells were treated with propolis at a concentration of4,8,16,25μg/mL.Conclusion:1. The secretion of IL-8, TNF-a, IL-10and NO were stimulated by propolis.2. Propolis downregulated the iNOS, PTGS-2and IL-8expressions in DNA transcription level, RNA level and protein level in Hela cells.3. The Western blot and RT-PCR analyses revealed that the expression of Egrl and p21in the Hela cells was significantly inhanced by higher concentrations of propolis, and the mRNA level of Ki67was attenuated, indicating that these factors may be involved in cellular proliferation or tumor inhibition.
