Objective:1.The aim of this study was to assess the association between endothelial nitric oxide synthase (eNOS) gene-786T/C and894G/T polymorphisms and acute coronary syndrome (ACS) in Xinjiang Uygur and Han.2. Comparison of the-786T/C and894G/T gene polymorphisms of eNOS gene and whether differences exist between Xinjiang Uygur and Han.Methods:1. We selected294ACS patients and147controls from the Uygur population,280ACS patients and195controls from the Han population, all of whom were in hospital in the cardiovascular department of Xinjiang Uygur Autonomous Region People’s Hospital from Jul.2011to Jul.2013.2. We Used the ABI7900HT fast real-time fluorescent quantitative PCR instrument to study-786T/C,894G/T gene polymorphisms of all cases.3. We Used SPSS17.0statistical software package for statistical processing, χ2test, t test, single factor analysis of variance, multivariate logistic regression analysis to calculate relative risk and their95%confidence interval.Results:1. For the-786T/C polymorphism, there was no significant difference in the frequencies of the genotypes between ACS cases and controls in Uygur (P=0.336). There was significant difference in the frequencies of the alleles between ACS cases and controls in Uygur (^=0.038). There was significant difference in the frequencies of the genotypes between ACS cases and controls in Han (P<0.001). However, The C allele frequency was significantly higher in ACS cases than that in controls in Han (P<0.001), Logistic regression analysis revealed that the risk of ACS was2.701times higher in individuals carrying CC+CT genotype than that in individuals carrying TT genotype (OR=2.701,95%CI=1.326-4.173, P <0.01).2. For the894G/T polymorphism, there was no significant difference in the frequencies of the genotypes between ACS cases and controls in Uygur (P=0.689). There was no significant difference in the frequencies of the alleles between ACS cases and controls in Uygur (P=0.859). There was significant difference in the frequencies of the genotypes between ACS cases and controls in Han (P=0.007). There was no significant difference in the frequencies of the genotypes between ACS cases and controls in Han (P=0.777). There was no significant difference in the frequencies of the genotypes and the alleles between Uygur and Han (P=0.077and P=0.444).3. Compared with the control group, the level of serum vWF in the acute coronary syndrome group patients was significantly increased(P<0.01). In the acute coronary syndrome group, the levels of serum vWF was positively correlated with systolic blood pressure, diastolic blood pressure, triacylglycerol, Low density lipoprotein cholesterol and fasting blood glucose (r=0.708,0.669,0.756,0.878,0.883;P<0.01). Multiple linear stepwise regression analysis revealed that diastolic blood pressure, triacylglycerol, Low density lipoprotein cholesterol and fasting blood glucose were the independent determinants of vWF levels (P<0.01).Conclusion:l.The eNOS gene may be associated with ACS in Xinjiang Han, but not associated with ACS in Xinjiang Uygur. However, for the-786T/C polymorphism, the C allele frequency was significantly higher in ACS cases than that in controls in Xinjiang Uygur (P=0.038).2. The levels of serum vWF was increased in acute coronary syndrome group and there are certain correlations between serum vWF and blood pressure^blood lipids and fasting blood glucose.
