Expression And Significance Of PTEN、ki-67and P53in Endometrial Carcinoma

Abstract/Summary:
ObjectiveTo investigate the expression and clinical significance of PTEN, Ki-67and P53innormal endometrium,simple hyperplasia,complex hyperplasia and endometrial carcinoma,to explore the association between PTEN, Ki-67, P53and endometrial carcinoma.MethodsA total of88formalin-fixed, parraffin-embedded tissue specimens used in this studyand selected from the Department of Pathology, women and Children Health Hospital ofjinan, including13cases of normal endometrium,28cases of simple hyperplasia,14casesof complex hyperplasia and33cases of endometrial carcinoma,were used in this study.Theexpression of PTEN、Ki-67and P53were examined by the SP method of immunochemistry,using SPSS19.0for statistical analysis and processing of the date, then p<0.05wasconsidered statistically significant.Results1.PTEN was mainly localized in cell cytosol,only a tiny minority localized in nucleus,orange particles.Significant differences of PTEN expression were noted between thenormal endometrium(positive rate:100%), simple hyperplasia(positive rate:83.3%),complex hyperplasia(positive rate:38.5%) and endometrial carcinoma(positiverate:36.4%)(p<0.05). The simple hyperplasia and the normal endometrium(p>0.05), theendometrial carcinoma and the complex hyperplasia group(p>0.05), there was no staticallysignificant. The positive expression rate of normal endometrium group was higher thancomplex hyperplasia and the difference was statically significant(p<0.01), the positiveexpression rate of normal endometrium group was higher than endometrial carcinomagroup(p<0.01), the positive expression rate of simple hyperplasia group was higher thancomplex hyperplasia group(0.01<p<0.05), the positive expression rate of simplehyperplasia group was higher than endometrial carcinoma group and the difference was statically significant(p<0.01). These results revealed a negative correlation between PTENexpression and the severity of endometrial diseases(r=-0.499**). PTEN expression closelyrelated to the tissue classification, clinical stage, myometrial invasion and the lymph nodemetastasis of endometrial carcinoma.and an obvious decresased expression was observedin the specimens with lower tissue differentiation, higher clinical stage, deep myometrialinvasion and positive lymph node metastasis of endometrial carcinoma.2.Ki-67was localized in nucleus,brown particles. In normal endometrium, simplehyperplasia, complex hyperplasia and endometrial carcinoma, the positive expression rateof Ki-67were7.7%,14.3%,30.1%,60.1%respectively. Statistical significance was onlyobserved between the normal endometrium and the endometrial carcinoma group, thesimple hyperplasia and the endometrial carcinoma group. These results revealed a positivecorrelation between Ki-67expression and the severity of endometrial diseases(r=0.462**).Ki-67expression closely related to the tissue classification of endometrial carcinoma, andan obvious increase expression was observed in the specimens with lower tissuedifferentiation of endometrial carcinoma.No correlation was observed between Ki-67expression and the clinical stage of patients with endometrial carcinoma, myometrialinvasion and metastasis.3.P53was localized in nucleus,and was brown particles.In normal endometrium,simple hyperplasia, complex hyperplasia and endometrial carcinoma, the positiveexpression rate of P53were100%,89.3%,64.%,42.4%respectively.And Statisticalsignificance was observed between the normal endometrium and complex hyperplasiagroup, the simple hyperplasia and endometrial carcinoma group, the normal endometriumand endometrial carcinoma group. These results revealed a negative correlation betweenP53expression and the severity of endometrial diseases(r=-0.497**).P53expressionclosely related to the clinical stage of endometrial carcinoma,and an obvious increaseexpression was observed in the specimens with higher clinical stage,No correlation wasobserved between P53expression and the tissue classification of patients with endometrialcarcinoma, myometrial invasion and metastasis.Conclusion1.In normal endometrium, simple hyperplasia, complex hyperplasia and endometrialcarcinoma,positive expression of PTEN decreased with the increased degrees ofendometrial diseases,it negatively correlated with the degrees of endometrial diseases.PTEN expression negatively correlated with the tissue classification, clinical stage,myometrial invasion and the lymph node metastasis of endometrial carcinoma. 2.With the degree of endometrial lesions deepened, the positive rate of Ki-67expression increased. Ki-67expression closely related to the tissue classification ofendometrial carcinoma.3.With the degree of endometrial lesions deepened,the positive rate of P53expressiondecreased. P53expression closely related to the clinical stage of endometrial carcinoma.
