ObjectiveBy studying the proliferation and angiogenesis abilities of bone marrow mesenchymalstem cells from pulmonary hypertension rat and investigating their biologicalcharacteristics,to provide preliminary experimental evidence for the treatment ofpulmonary hypertension by transplanting BMSCs.MethodsThe PAH model was made By injecting10%monocrotaline injection (60mg/kg) intorat intraperitoneal, the normal group was reproduced by injection of correspondingnormal saline according to the weight; After3weeks of injecting monocrotaline,hemodynamic parameters (mean pulmonary artery pressure) and the quality of rightventricle, interventricular septum and left ventricle were measured, then the index of theright ventricular hypertrophy was calculated by RV/(LV+S); The strcture of pulmonaryarterioles was observed by HE staining; blood analysis done on rats measured thepercentage of blood neutrophils; density gradient centrifugation and adherent cultureisolation and culture of PAH and normal rats BMSCs, BMSCs morphology was watchedunder a microscope; BMSCs surface antigen identification was made by flowcytometry,and angiogenesis experiments were done in vitro; BMSCs growth curves wasmade by CCK-8assay.Results1,Three weeks after monocrotaline injection,mPAP and RVHI were increasednotably (P<0.01)in PAH rats compared with normal rats.2, Compared with normal group,the percentage of neutrophils in PAH rats was markedly increased (P<0.01).3, HEstaining showed pulmonary arterioles of rats PAH group was significantly thicker thannormal group.4, The rat BMSCs surface markers were detected by flow cytometry:compared with normal rats, PAH group rat BMSCs positive surface markers (such asCD44, CD90) expression was markedly lower (P<0.05), while negative surface markers (such as CD31, CD34, CD45)expression was notably increased (P<0.05).5, Theproliferation ability of BMSCs PAH group rat was significantly reduced than the normalgroup.6, Angiogenesis experiments: rat BMSCs compared with the normal group, thenumber of BMSCs blood vessel formation cavities was significantly reduced(P<0.01) inPAH rats.Conclusion1, The model of pulmonary hypertension was successfully made by injectingmonocrotaline into rat intraperitoneal2,Inflammation was presented in PAH rats.3, Bonemarrow mesenchymal stem cells were acquired by Ficoll density gradient centrifugationand adherence screening, but the purity of PAH rats BMSCs was lower than normal group.4, The ability of BMSCs growth, proliferation and angiogenesis was decreased in PAHgroup.
