Purpose:To investigate effect of bone marrow stromal cells (MSCs) and intravenous injection on angiogenesis in infracted region of rabbit.Material and method:Models of acute myocardial infarction were established by ligating left anterior descending coronary artery on big-eared Japanese rabbits. They were divided into three groups which are model control group,point injection stem cell group and intravenous injection of stem cell group.After 1 week, suspension of MSCs by isolating,culturing amplifying and labeling injected in point injection stem cell group and intravenous injection of stem cell group was 1×l06 cells /point and 2×l06 cells / vein respectively. After 5 weeks,in order to observe whether MSCs in infracted region were transfer or not,Brdu immunohistochemical staines method was adopted. Effect of MSCs injected into points and vein on angiogenesis in infracted region were observed by using immunohistochemical method to investigate CD31 of infracted cardiac muscle tissue. Mechanism of MSCs injected into points and vein in infracted region were detected by using immunohistochemical method to investigate bFGF. Rabbit mortality rate of three groups were obser-ved.Results:1,Brdu and cTnT stains were both positive in point injection stem cell group and intravenous injection of stem cell group and negative in model control group and point injection stem cell group .2,Compared with normal group,The mean OD of CD31 in infracted region in point injection stem cell group were low(P<0.01), intravenous injection of stem cell group was high (P>0.05);Compared with point injection stem cell group, The mean OD of CD31 in model control group was low(P<0.05);Compared with model control group,intravenous injection of stem cell group was high(P<0.01).3, Compared with normal group ,the mean OD of BFGF in infracted regionin model control group and point injection stem cell group was high(P<0.05),and intravenous injection of stem cell group was low(P<0.01); Compared with point injection stem cell group,(P<0.05)and intravenous injection of stem cell group (P<0.01) were both low. Compared with model control group, intravenous injection of stem cell group was low(P<0.01).Conclusion:1,Stem cell injected into points and veins can transfer into infracted region And can differentiated into myocardial-like cells.2,Expression of CD31 in point injection stem cell group and intravenous injection of stem cell group increased and angiogenesis in infracted region.3,Angiogenesis mechanism of MSC injected into point was MSC stimulate expression of bFGF in infracted region.4,There is no significant difference of Angiogenesis in infracted region between point injection stem cell group and intravenous injection of stem cell group.
