ObjectivesExpression of the outcome of Anterior Cruciate Ligament Reconstruction withaitificial ligamnent made by sik and angiogenic effect and biomechanical effect of aitificialligamnet made by silk treated by Plasma rich in Platelt in ACL reconstruction at earlystage.Methods32health adult New Zealand white rabbits were removed two sides of ACL. Aitificialligament made by silk treated by PRP was transplanted into one side knee to substitute theoriginal ACL,while the other side only treated by saline as control.4rabbits were classifiedas sham group. The grafts were harvested4,8,16,24week from the32rabbits, respectively.The histological examination was made after4,8,16,24weeks of operation. Theintravascular injection of ink was used to observe the revascularization of the graft.Thespecimens were stained with HE and Masson method to observe the histological changesof the graft. The complex of femur-graft-tibia was biomechanical analysis.ResultsThe implanted was gradually covered by collgen fiber after the surgery and new bloodvessels and cells gradully invaded into it. The implanted was similar to the normal ACL at24weeks postoperative. The new born cells exhibited fibroblast morphology and weredistributed along the direction of mechanical loads.When compared with PRP at same time,the new blood vessel and new born cells of group PRP were significant higher than that ofgroup NS (P<0.01). New blood vessel of group PRP were significant higher than that ofgroup NS (P<0.01). Mew blood vessel of group PRP and NS was505±22,1214±84,619±23and402±27,823±38,521±30at4,8,16week postoperative,respectively(P<0.01). The timeof neovascular fomation and the depth of vascular penetration into the graft of the groupPRP are superior to those in the group of NS. New cells of group PRP and NS was261.25±12.42,386.50±36.57,276.50±34.07and198.25±6.55,304.25±52.64,206.00±16.08 at4,8,16week postoperative,respectively(P<0.01).The bew vessel and cells of group PRPand NS was330±22,309±32and186.25±21.93,175.00±13.69at24week postoperative,respecyively, there was no significant diference between them (P>0.05). The maximumload of group PRP and group NS was99.93±7.45N,90.64±5.37N,49.25±2.89N,49.40±1.84N and83.32±6.14N,53.77±9.24N,18.63±2.09N,13.53±1.65N at4,8,16,24week posterative,respectively(P<0.01). The tensile strength of regenerated ligament ofgroup PRP and group NS was decrasace with time, but the rensile atrength of regeneratedligament of group PRP can met the mechanical repuirements.ConclusionAfter ACL reconstrucyion used artificial ligament made with silk,the silk artificialligament can be converted to a ligament tissue similar to rabbit anterioi cruciate ligamnetin the rabbit knee joint cavity.Platelet rich plasma is beneficial for the earlyrevascularization and fibroblasts growing of the graft,and it can obviously decress thebiomechanics losses of newborn ligament tissue. PRP could be promoted on thereconstruction of ACL.
