2010Chinese pharmacopoeia have been recorded:Banlangen is usually used Traditional Chinese Medicine (TCM) being valued to have antiviral, antibacterial, antiendotoxic, immune regulatory activities and traditionally used for treatmenting seasonal febrile diseases, mumps, pestilence, eruptive diseases, inflammatory diseases with redness of skin, and sore throat.This dissertation consists of three parts, which belong to the chemical constituents of the root of Isatidis indigotica. The studies are as follows:In first part, the review about chemical components and pharmacological activities of BanLanGen is summarized, through consulting literatures at home and abroad. The research approach was carried out by analyzing present state of the BanLanGen research.In second part, the70%EtOH extract from the root of Isatidis indigotica Fort was concentrated to give a residue. The residue was separated by column chromatography on silica gel, preparative thin layer chromatography and high performance liquid chromatography (HPLC) to give11compounds. By means of IR spectra,’HNMR spectra,13CNMR spectra and Mass spectra, the ten chemical structures of these compounds are determined. The known compounds were identified by spectral analysis and compared with those reported in literatures or from authentic samples. Among these compounds,3isolated for the first time from Isatidis indigotica Fort are elucidated.In third part, the major compounds skeleton of BanLanGen were analysed, the possible biosynthetic pathways were discussed and the main compounds of spectrum rule was summed up on the basic of the compounds afforded by our group.At last, antioxidation of different extracts from Isatidis indigotica Fort were studied in viro to supply the separating base of antioxidative components. The extracts were prepared by macroporous resin and anion/cation exchange resin. The three compounds was isolated by our group. Their antioxidant activities were assessed by two complementary test systems, namely l,l-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity and ferric reducing antioxidant power (FRAP) assay. At a Result, the total lignin displayed strong activity in all test systems.(DPPH IC50:160.6mg·L-1; FRAP:0.78mmol·g-1) Banlangen Keli (DPPH IC50:595.97mg·L-1; FRAP:0.12mmol·g-1) showed relatively lower activity. Total organic acids and total alkaloid have a little activity. Among the tested compounds, considerable activity of isolariciresinol was observed in all test systems (DPPH IC50:24.29μM; FRAP:7.16mmol·g-1). Antioxidative components could be justified existing in the total lignin initially.
