Studies On The Secondary Metabolites Of Verrucosispora Sp.NS0172 And Streptomyces Sp.S035

Ansamycin is a class of macrolactam antibiotics with unique structure and diverse biological activities,it is mainly present in actinomycetes and a few plants.Gene sequencing and bioinformatics analysis showed that actinomycetes have huge secondary metabolic potential.After perform some biological genetic manipulation on the gene,we get overexpressed mutants,then we would obtain target compounds,it will play a major role in the study of new drugs.In this paper,the mutant strains of NS0172 and S035 are used as experimental materials,then study their secondary meta'bolite,a total of 30 compounds were isolated,and the structures of compounds were determined by MS?NMR,of which 20 were new compounds,including 1 triketide and 12 tetraketides ansamycins.Verrucosispora sp.NS0172 was isolated from Xiamen.PCR sequencing showed that NS0172 was an AHBA positive strain,it has the potential to produce ansamycin compounds.Our team's Master Li Wen has overexpressde vas 10 and vas 12,and got two mutant strains NS0172-OELAL and NS0172-SARP.We obtained 1 known compounds from the NS0172-OELAL;and 10 compounds were obtained from the NS0172-SARP,including 5 new compounds,a new triketide ansamycin and two new tetraketide ansamycins,compounds 2-4 were tested for antibacterial activity,but none of the compounds showed activity.Streptomyces sp.S035 is also an AHBA-positive strain.Gene sequencing and bioinformatics analysis showed that it can encode a novel carbon ketone with a novel pentaketide antamycin.Our team's master Wang Zishen constructed some mutant strains.The author selected three of them to study their secondary metabolites.First 10 new tetraketine ansamycins were obtained from the S035-LAL1,but none of the compounds showed activity;then 6 known pentaketide ansamycins were obtained from the S035-SARP;finally,3 new compounds were obtained from S035-LAL2,however,without pentaketide ansamycins was got.
