Objectives1.To estimate the function of the Polygoni Multiflori Preparata on hematopoietic system, the effect of Polygonum multiflorum main component on proliferation of hematopoietic progenitor cell were researched.2. To lay the foundation for the further research on Polygoni Multiflori Preparata, the effect of the different blood sampling time point containing serum on the proliferation of hematopoietic progenitor cells were researched.3.To explore the molecular mechanism of the Polygoni Multiflori Preparata in enrich the blood, the effect of Polygonum multiflorum on the apoptosis of bone marrow cells, cycle and adhesion molecule expression, the tyrosine phosphorylation expression of red cell membrane band3protein were researched.Methods1. Observe the effect of the raw Radix Polygoni Multiflori, Polygoni Multiflori Preparata high dose、medium dose、low dose and its main monomer composition on the hematopoietic system of the anemic rats in vivo.2. Observe the effect of the raw Radix Polygoni Multiflori, Polygoni Multiflori Preparata and its main monomer composition on the proliferation of hematopoietic progenitor cells in vitro semi-solid cell culture.3. Intragastric administration untreated rats the Polygoni Multiflori Preparata,4days of dosing, after the last administration,5min,15min,30min,45min,1h,1.5h,2h,4h,6h,8h,10h and24h containing serum was collected. Respectively, to study the effect of ethanol precipitation containing serum and untreated containing serum on the proliferation of hematopoietic progenitor cells.4. Analysis the expression of deficient model rat bone marrow cell adhesion molecules of CD44CD54and red cell membrane adhesion molecule CD35, by flow cytometry after continuous intragastric administration Polygonum multiflorum of9days.5.Study the effect of Polygonum multiflorum on apoptosis of bone marrow cells and cycle in deficient model rat after continuous intragastric administration of9days.6. Using western blot techniques, study the effect of Polygonum multiflorum on the expression levels of tyrosine phosphorylation of the red cell membrane band3protein.Results1. In vivo, the raw Radix Polygoni Multiflori and Polygoni Multiflori Preparata low dose (1g/ml) should not improve the function of the deficient model rats hematopoietic systen. Polygoni Multiflori Preparata medium dose(2g/ml)、high dose(4g/ml) improve the hematopoietic function, exceptionally, the most obvious improvement is the medium dose.2. The raw Radix Polygoni Mul tiflori and Polygoni Mul tiflori Preparata should not promote the growth of CFU-GM cells. Monomer components physcion, emodin, emodin monomethyl ether-8-0-beta-D-glucoside and emodin-8-0-β-D-glucoside can promote the proliferation of CFU-GM cells, and the lower the concentration of monomer components, the more obvious effect.3. The Polygonum Mul tiflori Preparata containing serum can improve the growth of CFU-GM cells, and the raw Radix Polygoni Multiflori containing serum could not.With ethanol precipitation, the Polygonum Multiflori Preparata containing serum of1.5h has the most obviously effect on CFU-GM cell proliferation, and1h without any treatment.4. The Polygoni Multiflori Preparata can significantly reduce the high expression of the bone marrow cell adhesion molecule CD54and red cell adhesion molecules CD35because of anemia.5. The raw Radix Polygoni Multiflori and Polygoni Multiflori Preparata can promote the anemic rat bone marrow cells from G1phase to S phase.6. The raw Radix Polygoni Multiflori and Polygoni Multiflori Preparata can enhance the expression of tyrosine phosphorylation of erythrocyte membrane band3protein in deficient model.ConelusionThe Polygoni Multiflori Preparata can improve the hematopoietic function of anemic rat, might by promoting the proliferation of hematopoietic stem cells CFU-GM, regulating the expression of adhesion molecules of bone marrow cells and red cell membrane, accelerating the bone marrow cells from G1phase to S phase,repairing damaged red blood cells.
