The Research Of Effective Chemicals Of Taxillus Chinensis Danser And Channel Tropism Based On Disease-Effect-Bioanalysis

Objective:We observed effect of Taxillus in rheumatoid rats,osteoporotic rats and investigated effective chemicals of Taxillus,find the relevance between body distribution and channel tropism. Methods:1. Rats were divided into normal group,model group,Positive group,Taxillus different dose groups.After 8 days of rats except normal induced by Freunds complete adjuvant,gave normal and model group saline,Positive group Glucosidorum Tripterygll Totorum,Taxillus different dose groups different doses of water solution of Taxillus for 21 days. During the period of medication,measured the diameter of the left ankle of rats every 7days.Take blood of all rats and measure Interleukin-1β,Tumor necrosis factor alpha.Measure the levels of rutin, avicμlarin and quercetin in tissues of Taxillus high dose rats by HPLC. Use DAS2.0 software to deal with Pharmacokinetic parameters.2. we gave rats exceppt normal group with retinoic acid 70 mg/kg every morning for 14 days,gave normal and model group saline,Positive group Longmu Zhuanggu granμles,Taxillus different dose groups different doses of water solution of Taxillus every afternoon for 28 days.Take blood of all rats and measure osteocalcin, alkaline phosphatase.Take the left femur of all rats and measure weight, length. Measure the levels of rutin, avicμlarin and quercetin in tissues of Taxillus high dose rats by HPLC. Use DAS2.0 software to deal with Pharmacokinetic parameters. Resμlts:1. Water solution of Taxillus showed positive influence in Rheumatoid rats,and had a tendency to drop IL-1β、TNF-α.Pharmacokinetic data showed that rutin has a small amount of the distribution in stomach,avicμlarin AUC0-t was stomach>small intestine>kidney,the body maintain time was kidney=stomach>small intestine>lung>heart.Quercetin AUC0-t was stomach>kidney>liver>heart>lung>spleen>small intestine>brain>large intestine>blood, the body maintain time was stomach=kidney=liver=heart=lung=spleen=small intestine=brain> large intestine>blood.2. Water solution of Taxillus had positive influence in W,W/L,W/L of osteoporotic rats,and reduced the OT,ALP content.Pharmacokinetic data was as follows.Rutin was Only in stomach. Avicμlarin AUC0-t was stomach>kidney>brain>small intestine>large intestine> liver>lung, the body maintain time was stomach=kidney>brain>large intestine>small intestine=spleen>liver=heart=lung>blood.Quercetin AUC0-twas kidney>stomach>liver>heart>Lung>spleen>small intestine>large intestine> brain, the body maintain time was kidney=Stomach=liver=heart=lung=spleen =brain >large intestine>small intestine> blood. Conclusion:Water solution of Taxillus has obvious effect in in rheumatoid rats and osteoporotic rats,avicμlarin and quercetin are effective chemicals of Taxillus. Avicμlarin and quercetin in distribution in rats have a high consistency in Taxillus’ channel tropism, avicμlarin and quercetin have high correlation in Taxillus’ ingredients.
