ObjectiveTo study the effects of astragaloside IV on the protein expression of Bcl-2、Bax、and caspase-3 in expeirmental myocardial ischemic-reperfusion rats, and to investigate its mechanism.Methods40 SD rats were randomly divided into the sham operation group, ischemia-reperfusion group,Ischemia precondition group and astragaloside IV group, 10 rats in each group(5 rats will give westernblotting text). Each rat had been record Ⅱ electrocardiographic(ECG) before the experiment. astragaloside IV group was given common feed +Astragaloside IV 10mg/(Kg·d),then saline were given to the other groups at 10 ml/kg weight; All of these medicines were given to rats at 10 ml/kg weight and one time each day. The administration would continue 7 days. Until the 8th day, Ischemia precondition group give 3 times ligations, gap 30 s each time,sham operation group without ligation,The remaining 2 groups with left anterior descending coronary artery ligation method of copying models of myocardial ischemia, 2 hours reperfusion after the release the ligature 30 min, After 2 hours all of the rats were anaesthetized. The rats were killed, HE staining was employed to analyze the pathological damage in myocardium of heart apex and Using DNA in situ end labeling(TUNEL) and western blotting localization of the apoptosis of myocardial cells and immune staining to detect the protein expression of Bcl-2、Bax and Caspase-3.Results1、Pathological changes: The sham operation group has no significant histological changes; Ischemia-reperfusion group cardiac dilatation a n d c o n g e s t i o n o f c a p i l l a r i e s b e t w e e n t h e f i b e r s, w i t h f o c a l i n f i l t r a t i o n o f i n f l a m m a t o r y c e l l s, c e l l s w e r e s e v e r e l y e d e m a e d, states were unclear; The myocardial fibers of the high and low dose groups of astragaloside IV and isosorbide dinitrate group were mild swelling, showing cell boundaries clearer.2. The comparison of average absorbancy of Bax、Bcl-2 and caspase-3 in different rats groups realized by the comparison of grey level of positive cells in method of Westernblots. the result is proved that: the expression of Bax、Bcl-2 and caspase-3 protein of cardiac muscle tissue in four groups have significant difference(P <0.05), by being compared between any two groups further.3.The expression of Bax、Bcl-2 and caspase-3 protein in model group and the astragaloside IV group is prominently higher than that of sham-operation group, Bax 、 caspase-3 in model group is higher than astragaloside IV group(P<0.05) while lowerin the matter of Bcl-2(P<0.05).The preconditioning group is indiscriminating as the astragaloside IV group. The difference show the significance of statistics.Conclusions1 The astragaloside IV is effective on reducing the cell apoptosis obviously in the process of myocardial ischemia reperfusion of rats;2 The astragaloside IV protect the myocardial cell by adding the expression of Bcl-2,Caspase-3 and reducing the expression of Bax;3 The effect of astragaloside IV on protecting myocardial ischemia reperfusion injury for rats is not discriminating as the recognized effect of ischemic preconditioning.
