Objective:To develop a new gene microarray on the basic of ionizing radiation inducible genes, confirm the transcriptional profiles induced by X-ray irradiation to explore the different biological mechanisms of ionizing radiation, explore the molecular mechanism of different radiation doses and optimize microarray as the biological dosimeter.Methods:1) A new microarray was delevopled on the background of 50 ionizing radiation inducible genes.2) Human lymphocytes were irradiated by X-ray with radiological doses of 0.1Gy,0.5Gy,2Gy.3) After 4 h and 20h, Microarray was used to analyze the gene expression changes of the lymphocytes. The genes which threshold was Fold Change= 1.5 were identified as radiation inducible genes through NimbleScan software.4) GO analysis method were used to analyse the biological function of some parts of radiation inducible genes which sreened out.Results:1) The up-regulated genes at 4h after irradiation of 0.1 Gy is XPC, down-regulated is SSBP2, The up-regulated genes at 4h after irradiation of 0.5Gy is BBC3,GADD45A,IER5,XPC, down-regulated is CCT5,SSBP2, The up-regulated genes at 4h after irradiation of 2Gy is,BBC3,CDKN1A,GADD45A,IER5,TP53IP3,XRCC5,XPC, down-regulated is CCT5,SSBP2, The up-regulated genes at 20h after irradiation of 0.5Gy is BAX,BBC3,TP53I3,XPC, down-regulated is CCT5,PCNA, The up-regulated genes at20h after irradiation of 2Gy is BBC3,CDKN1A,TP53IP3,XRCC5, XPC, down-regulated is CCT5,PCNA.2) The predominant functional groups responding to low-dose radiation are those involved in cell signaling, signal transduction, cell/tissue defense, homeostasis. And the responding genes induced by high dose irradiation involved in apoptosis, cell proliferation and cell cycle arrest.Conclusion:1. Microarray on the basic of ionizing radiation inducible genes is a powerful tool as biological dosimeter.2. BBC3, CCT5,CDKN1 A, GADD45A,IER5, PCNA can be used as a radiation biological dosimeter.3.DDB2 expressed different results in different experiments, whether it can be used as a radiation biological dosimeter requires careful conxideration.
