Objective: To investigate the changes of intestinal defensins in rat with NSAIDs related enteropathy and it’s influence on local intestinal inflammation. Methods: Thirty-four SD adult rats, male and female each half.They were randomly and equally divided into experimental group and control group.The experimental group was taken aspirin with 100 mg / kg by intraperitoneal injection twice a day, and anther 17 rats in control group were injected of normal saline twice a day. About two weeks later,two groups were fasted for 24 hours, forbidden to drink eight hours,fixed on the operating panel, disinfected, opened the abdominal cavity,we took the portal vein blood,Abdominal aorta blood about 2 ml separately. Then we found the ileocecal valve and cut ileum about 2cm at the proximal end of about 5cm.The intestinal tissue were divided into two equal parts, one for doing tissue homogenates, one for paraffin sections.The levels of RD5 and RD6 were detected by Immunohistochemistry methods. IOD values were measured by image analysis software. The levels of IL-1β, IL-8, IL-10, ANCA and endotoxin in the blood from the abdominal aorta and the portal vein were detected by ELISA. Results: The mucosal of the experimental group cloud be seen edema, scattered bleeding and erosionhe,but the control group was not. IOD values of RD5,RD6 in the experimental group were much higher than those in the control group(p<0.01),mainly expressed in the midgut gland basal crypt cells, and peripheral goblet cells also stained. IL-1β,IL-8,IL-10,ANCA in experimental group were also higher than those in the control group,too(p<0.01). There was no statistical significance about the levels of endotoxin in serum from the abdominal aorta and the portal vein between the experimental group and the control group(p>0.01). Conclusions: The mucosal injury did not reached the degree of bacteria and endotoxin translocation by intraperitoneal injection of 100mg/kg aspirin for two weeks. Immune barrier and local inflammatory response migth play an important role in the occurrence and development of NSAIDs related enteropathy. Autoimmunity might participate into the mechanism of NSAIDs related enteropathy at the same time,and in which,the alpha-defensins might play multiple roles in the process.
