Effect Of Yiqi Jiedu Decoction On Tumor Bearing Mice Hepatoma H22 Cell Proliferation In Vivo

Purpose: A randomized controlled principle,according to the theory of traditional Chinese medicine,after the appropriate compatibility,to explore the effects of Yiqijiedu compound on the growth of tumor in mice bearing H22 tumor cells,the experimental basis for the anti- liver cancer of Chinese traditional Chinese Medicine。Method : The mice hepatoma H22 cells were inoculated with the right axillary subcutaneous under the aseptic conditions,each 0.2ml(1×107个/ml),24 hours after randomization,7 days after observation inoculation effect。Randomly grouping mice that have successfully grown tumors,10 rats in each group,half male and half female,the liver cancer model group(model group)、chemotherapy group(group 5-FU)、effect of Yiqi Jiedu Decoction group(high, low, high, low, in the YJP group),another 10 mice were used as normal control group。On the next day,5-FU group received intraperitoneal injection of 5-FU 10mg/kg, administered 1 times every other day;YJP takes Yiqi Jiedu Decoction with 40, 20, 10ml/kg dose gavage,1 times a day;The model group was orally at the dose of saline 20ml/kg daily;Normal control group was given fresh water, and did not do any other treatment。A total of 12 mice were observed,daily weighing animal weight,observe the color、eating、activity and death of the animals in each group;The following day after the last Administration,all animal were killed by cervical dislocation,the blood test, peripheral blood tumor inhibition rate, spleen index were statistically analyzed;Paraffin embedding and pathological sections of the tumor tissue were carried out,the expression of Bcl-2 protein was observed by immunohistochemistry 。 The method of the side groups of the Qi and detoxification party is the same as above。Result: After treatment with Yiqi Jiedu Decoction,The rise of spleen coefficient of the rats,The white blood cell count increased,The tumor volume decreased,Cell smear under light microscope, cell proliferation decreased;The results of immunohistochemistry showed that the expression on hepatocellular carcinoma cells H22 Bcl-2 reduction。The above results were statistically significant。Conclusion:Effect of Yiqi Jiedu Decoction can prolong the survival time of tumor bearing mice;Effect of Yiqi Jiedu Decoction can inhibit the proliferation of H22 cells in vivo,that is the inhibition of cell proliferation plays an important role in the way of treatment in this experiment;Effect of Yiqi Jiedu Decoction can inhibit the expression of Bcl-2 in hepatocellular carcinoma H22 cells,it is suggested that the expression of the cell apoptosis factor can inhibit the cell proliferation by regulating the expression of apoptosis factor。...
