Pur Pose:Objective: Through the observation of effect on the lung cancer mouse Lewis’ Pathological morphology change and the protein expression of related factor apoptosis Caspase-3 by Added Qinghao Biejia Decoction made from Yiqi Yangyin Huayu method, we investigate the tumor inhibitory effect and mechanism of Lewis under the Yiqi Yangyin Huayu method.Method:A total of 60 male mouse C57BL/6J were randomly divided into six ghroups with 10 rats in each group, such as model group, low, medium and high dose of the added wormwood and turtle soup group, cisplatin group and comprehensive group(Chinese medicine with medium dose plus cisplatin).Give drug delivery treatment at the seventh day, and the animals were executed at the twenty-first day. General living status of experimental mice were observed and we also recorded the changes of mouse tumor volume, calculated the inhibition rate of tumor and lung metastasis inhibition rate, and through HE staining to observe the Pathological morphological changes and the immunohistochemistry of Caspase-3 protein expression. Through SPSS19.0 we conducted data statistics and analysis.Results:1. The tumor grew rapidly in 15 days. The average values of tumor volume in the model group compared to other groups were statistically significant(P<0.05 or P<0.01) at twenty-first day.2. The average tumor weight of the treatment group compared with the model group were no significantly different(P>0.05) in the low dose group, and the other groups were significantly different(P<0.01).Compared with cisplatin group, the dose of tumor in the traditional Chinese medicine dose group was lower than that in cisplatin group and statistically significant(P<0.05 or P<0.01).Compared to the Chinese medicine group with different doses and cisplatin group, the tumor weight comprehensive group of is lower than other drugs, with statistical significance(P<0.05 or P<0.01).The inhibition rate of TCM in Chinese group with low, middle, high dose, cisplatin group and combined group were 6.5%, 24.5%, 31.5%, 46.5%, 62%, respectively.3.There was no significant difference for Pulmonary surface metastasis nodules in the low dose of traditional Chinese medicine(P>0.05) compared with the treatment group and the model group. The other groups were significantly different(P<0.01). The inhibition rates of pulmonary metastasis in Chinese group with low, medium, high dose, cisplatin group and combined group were 11.83%, 27.96%, 33.33%, 37.85%, 44.09%, respectively.4. Under 400 times light microscope, the pathological changes of tumor were observed, the density of tumor cells decreased, the cells appeared death and apoptotic bodies, and the cells and pathological nuclei were decreased.5. With immunohistochemistry method, Caspase-3 Protein expression in tumor tissue were detected. Drug group could significantly promote the expression of Caspase-3 protein, and it was significantly different(P<0.01) compared with model group.Compared with Chinese group with different doses, the expression of Caspase-3 in cisplatin group was higher than that in traditional Chinese medicine, and it had significant difference(P<0.05 or P<0.01).Compared with Chinese group with low, medium, high dose and cisplatin group, the expression of Caspase-3 in the integrated group was higher than that in the other groups(P<0.05 or P<0.01).Conclusion:1. The added artemisia annua and turtle soup made from Chinese Yiqi Yangyin Huayu mechanism can obviously improve the survival state of lung cancer mice Lewis.2. The added wormwood and turtle soup made from Chinese Yiqi Yangyin Huayu mechanism can inhibit the mice tumor growth and lung surface transfer. It also has the effect of anti tumor, and synergistic effect combined with cisplatin chemotherapy drugs.3. The added wormwood and turtle soup made from Chinese Yiqi Yangyin Huayu mechanism can induce Lewis lung cancer cells apoptosis. The mechanism may promote the expression of Caspase-3 protein, and enhance it combined with cisplatin chemotherapy drugs.
