| Purpose: Observation of citrate complex kidney qi pill effect on the changes of TIMP-1,MMP-9 in serum and renal tissue of IgA nephropathy rats, to explore the mechanism of the drug in the treatment of IgA nephropathy.Method: A total of 80 healthy male Wistar rats of clean grade, weight(200±20) g, 1weeks after adaptive feeding, use the method of stratified random. The rats were divided into blank group 10, model group 70. The model for bovine serum albumin(BSA) oral gavage of25% carbon tetrachloride(CCL4) by subcutaneous injection of 0.25g/L solution and lipopolysaccharide(LPS) intravenous injection of established IgA nephropathy rat model, a total of 9 weeks. The validation of the model after the success of the model group rats were redivided into model group, control group, hematuria capsule, Qi Fu Shen pill high, medium and low dose group. Given the hematuria capsule control group was daily gavaged with hematuria capsule 0.547g/kg; give Gou Qi Fu Shen pill high dose group of 5.625g/kg·d-1by gavage, the middle dose group 2.813g/kg·d-1, intragastric administration of low dose group1.406g/kg·d-1 orally; the control group and the model group was given saline gavage daily,4ml/rat Total of 8 weeks of administration.The observation index:(1) The general state of rats;(2) Counting of rats in each phase of the red blood cells in urine and 24 hour urinary protein quantitative;(3) Teste d kidney function, liver function;(4) Were measured with ELISA method for the deter mination of serum and renal tissue MMP-9, TIMP-1;(5)Detection of IgA complex deposition by immunofluorescence method.Result:(1) During the experiment, rats were killed 19 in the blank control group, model group, 2rats died, 9 died, 8 were used to validate the model.(2) Better blank group rats during the whole experiment, no obvious abnormality. The model group rats were given subcutaneous injection of carbon tetrachloride showed varying degrees of depression, hair messy and not shiny, mouth nose secretions, diarrhea, or reduce the slow increase of body weight, urine,urine and other performance. After 2~3 weeks after treatment, the general state of rats in each treatment group improved, reducing urination, weight gain, which Gou Qi Fu dose groupimproved the most obvious in the kidney shot, the model group had no obvious change.(3)The normal group rats without hematuria and proteinuria positive performance, make urine red blood cell count model rats and 24 hour urinary protein quantitative increased significantly, after treatment decreased between two groups, the treatment group than in control group(P<0.05).(4) In serum and renal tissue of IgA nephropathy model rats decreased MMP-9, increased TIMP-1 in, After treatment, MMP-9 increased, TIMP-1 decreased, and the citrate complex kidney qi bolus dose group than in the control group hematuria capsule(P<0.05).(5) No significant difference was found between the groups of rats renal function and liver function(P>0.05),no statistical significance.Conclusion:(1) Methods using bovine serum albumin gavage and carbon tetrachloride, LPS injection could successfully establish animal model of IgA nephropathy.(2) Citric acid complex kidney qi pill can effectively reduce IgA nephropathy model rats24 hours urine red blood cell count and the reduction of 24 hours urinary protein excretion rate.(3) After testing compared with the control group, decreased the content of MMP-9 in serum and renal tissue of IgA nephropathy model rats, the MMP-9 was significantly increased after drug treatment was positively correlated with the disease; TIMP-1, serum and renal tissue of IgA nephropathy rats increased after drug treatment, the decrease of TIMP-1, and negatively correlated with the disease; that the abnormal expression of MMP-9, TIMP-1involved in the pathogenesis of IgA nephropathy.(4) We speculate that the citrate complex kidney qi pill is through the expression of MMP-9 and TIMP-1 regulation of IgA nephropathy rats serum and kidney tissue in quantity,can play a role in the treatment, improve the pathological state.(5) The pathological examination showed that citric acid complex kidney qi pill can effectively improve the IgA nephropathy rat model of renal pathology, disease and delay the further development, protect the kidney structure.(6) Through the comparison of the model rats with liver and kidney function of rats in the normal control group had no significant difference.(7) Therapeutic effect of citric acid complex kidney qi pill in reducing IgA, hematuriaand proteinuria, hematuria capsule is superior, and no damage of liver and kidney, is worth further research and promotion. |
