Objective To observe the effect of gemcitabine and cisplatin therapy on TXNIP expression in non-small cell lung cancer cells.Methods Non-small cell lung cancer patients were administrated with two course of gemcitabine and cisplatin, then the clinical specimens were collected before and after the treatment. The m RNA of TXNIP was detected by real time PCR, the protein were measured by immunohistochemistry and western blot. The acitivity of thioredoxin reductase(Trx R) was detected by colorimetry. The lung cancer cell line NCI-H1792 was cultured in vitro, and then was incubated by 0.2mg/L of gemcitabine and 0.5 mg/L cisplatin. After treatment, WST-1 was used to evaluate the proliferation of cells. Real time PCR and western blot were also selected to measure the expression of m RNA and TXNIP. Colorimetry was used to assess the enzyme activity of Trx R.Result 1) The m RNA of TXNIP in the healthy populations express to a certain level. In contrast, the TXNIP m RNA in NSCLC patients was lower than that in healthy control. After 2 two course of gemcitabine and cisplatin treatment, the TXNIP m RNA was gradually increased; 2) Immunohistochemistry demonstrated that in the lung of the healthy population, TXNIP was expressed to a certain level, while it decreased in NSCLC patients. Gemcitabine and cisplatin administration could increase its expression level. 3) The enzyme activity of Trx R was higher than that in healthy control(P<0.05), while Gemcitabine and cisplatin administration significantly inhibit its activity. 4) 0.2mg/L of Gemcitabine combined with 0.5mg/L cisplatin significantly decrease the proliferation of NCI-H1792 cells to 88%. As the concentration increased, the proliferation decreased gradually. 5) The m RNA of TXNIP was very low in NCI-H1792 cells. After different concentration of Gemcitabine and cisplatin treatment, the TXNIP m RNA and protein increased accordingly. 6) Gemcitabine and cisplatin treatment significantly decreased the enzyme activity of Trx R. When the highest concentration were used, the activity decreased to 43%.Conclusion 1.The combination of gemcitabine and cisplatin can upregulate TXNIP gene and protein expression in non small cell lung cancer patients.2.Gemcitabine combined with cisplatin can increase the expression level of TXNIP gene and protein in lung cancer cell line NCI-H1792 in vitro.
